TLR9信号通路与HPV16癌基因在宫颈病变中的作用研究

【摘要】 目的 研究旨在探讨TLR9、IRF7以及HPV16癌基因在宫颈病变进展中的作用及关系,为临床应用提供理论依据。方法 选取100例HPV16阳性患者为研究对象。随机分配为4个组别,每组25例。包括慢性宫颈炎组,宫颈上皮内瘤变Ⅰ级(CINⅠ)组,宫颈上皮内瘤变Ⅱ~Ⅲ级(CINⅡ~Ⅲ)组,宫颈鳞状细胞癌(SCC)组。采用免疫组织化学染色法检测宫颈组织中HPV E6和HPV E7蛋白的表达水平;采用Western Blot技术分析宫颈组织中TLR9、IRF7蛋白的表达水平;采用RT-qPCR检测宫颈组织中TLR9、IRF7 mRNA表达水平。结果 在慢性宫颈炎组、CINⅠ组、CINⅡ~Ⅲ组和SCC组的不同等级宫颈病变样本中,随着病变严重程度的增加,HPV E6与HPV E7蛋白于宫颈组织中的表达量以及染色综合评分表现出逐渐上升的趋势。TLR9与IRF7蛋白在宫颈组织内的表达水平呈现出递增趋势。TLR9与IRF7 mRNA在宫颈组织中的表达量显示出逐步上升的趋势,且上升变化幅度逐渐减小。差异均有统计学意义(P<0.05)。结论 本研究探讨TLR9、IRF7以及HPV16癌基因在宫颈病变进展中的作用。结果表明随着宫颈病变严重程度的加剧,TLR9、IRF7和HPV E6、HPV E7在宫颈组织内的表达水平呈现出递增趋势。揭示这些因子在宫颈病理变化中的重要性,表明宫颈病变的发展可能牵涉更为复杂的免疫调节机制。这不仅为宫颈病变分子机制的研究开辟了新方向,也为临床干预策略提供了理论支持。

【Abstract】 Objective The study aims to investigate the roles and relationships of TLR9, IRF7 and the HPV16 oncogene in the progression of cervical lesions, providing a theoretical foundation for clinical applications.Method A total of 100 HPV 16 positive patients will be selected as research participants and randomly assigned to four groups, with 25 patients in each group. The groups will consist of the following: the chronic cervicitis group, the cervical intraepithelial neoplasia grade I(CIN Ⅰ) group, the cervical intraepithelial neoplasia grade Ⅱ-Ⅲ(CINⅡ-Ⅲ) group, and the squamous cell carcinoma of the cervix(SCC) group. Immunohistochemical staining will be used to detect the expression levels of HPV E6 and HPV E7 proteins in cervical tissues. Additionally, Western blot analysis will assess the expression levels of TLR9 and IRF7 proteins in the same tissues. Finally, the mRNA expression levels of TLR9 and IRF7 will be measured in cervical tissues using RT-qPCR.Results In samples of cervical lesions from different groups-chronic cervicitis, CINⅠ, CINⅡ-Ⅲ, and squamous cell carcinoma(SCC)-the expression levels of HPV E6 and E7 proteins showed a gradual increase corresponding to the severity of the lesions. Similarly, the expression levels of TLR9 and IRF7 proteins in cervical tissues also exhibited an increasing trend. Furthermore, the expression levels of TLR9 and IRF7 mRNA in cervical tissue increased gradually, although the magnitude of this increase diminished over time.(all statistically significant at P<0.05).Conclusion This study examined the roles of TLR9, IRF7, and HPV16 oncogenes in the progression of cervical lesions. The results indicate that as the severity of cervical lesions increases, the expression levels of TLR9, IRF7, and the HPV oncogenes E6 and E7 in cervical tissues also rise. This highlights the importance of these factors in cervical pathology and suggests that the development of cervical lesions may involve more complex immune regulatory mechanisms. These findings not only pave the way for further research into the molecular mechanisms underlying cervical lesions but also provide theoretical support for clinical intervention strategies.
