PUM1通过糖酵解途径影响胶质瘤细胞的增殖
PUM1 affects the proliferation of glioma cells through glycolytic pathways
【摘要】 目的 探究Pumilio RNA结合家族成员1 (PUM1)对胶质瘤U251和T98G细胞增殖能力的影响及机制。方法 使用siPUM1对U251及T98G胶质瘤细胞系进行转染,通过CCK-8实验以及EdU实验检测PUM1对U251和T98G的细胞活力以及增殖的影响,通过乳酸生成实验检测PUM1和糖酵解途径的关系。结果 PUM1在胶质瘤中的表达水平较正常人星形胶质细胞更高,且敲低PUM1后,相比于对照组,实验组胶质瘤细胞的增殖能力减弱,乳酸生成量减少。结论 PUM1可通过糖酵解途径促进胶质瘤细胞的增殖。
【Abstract】 Objective To explore the effect and mechanism of PUM1 on the proliferation ability of glioma U251 and T98G cells.Methods Using siPUM1 to transfect U251 and T98G glioma cell lines,the effects of PUM1 on cell viability and proliferation of U251 and T98G were detected through CCK-8 and EdU experiments,and the relationship between PUM1 and glycolytic pathways was detected through lactate generation experiments.Results The expression level of PUM1 in glioma is higher than that of normal human astrocytes,and knocking down PUM1 reduces the proliferation ability and lactate production of glioma cells in the experimental group compared to the control group.Conclusion PUM1 can promote the proliferation of glioma cells through the glycolytic pathway.
