铁死亡和氧化应激在多发性骨髓瘤中相互作用的研究进展
Crosstalk between Ferroptosis and Oxidative Stress in Multiple Myeloma
【摘要】 铁死亡(ferropotsis)是一种由铁依赖性脂质过氧化驱动的程序性细胞死亡方式(programmed cell death,PCD),在生物学过程和形态学特征上不同于任何已知的程序性细胞死亡。铁死亡与多种代谢通路相关,包括铁稳态、脂质代谢以及氧化代谢,涉及血液系统恶性肿瘤的发生发展及治疗。氧化应激是细胞内氧化和抗氧化物质失衡的结果,所产生的过量活性氧(reactive oxygen species,ROS)可导致一系列细胞功能紊乱,也是肿瘤发展的重要因素之一,但大量的ROS积累可以产生细胞毒性,亦可导致癌细胞死亡。铁死亡的发生涉及氧化代谢,ROS参与了铁死亡通路的调节,是铁死亡发生的关键步骤,而铁死亡的发生必然导致脂质的过氧化。本文系统总结了铁死亡和氧化应激在多发性骨髓瘤发生、发展及治疗中的作用,以期为多发性骨髓瘤的治疗及克服耐药性提供新的策略。
【Abstract】 Ferropotsis is a form of regulated cell death characterized by the iron-dependent accumulation of lipid hydroperoxides to lethal levels,which is distinct from any known programmed cell death in the biological processes and morphological characteristics.The ferroptosis is correlated with numerous metabolic pathways,including iron homeostasis,lipid metabolism,and redox homeostasis,associating with the occurrence and treatment of hematological malignancies.Oxidative stress is a state of imbalance between oxidation and antioxidation,producing excessive levels of ROS,which can lead to deranged control of cellular functions and is also an important factor in tumor development.Ferroptosis pathway is associated with oxidative metabolism,and ROS not only participates in the regulation of ferroptosis and aslo play a key role in it.Whereas,the occurrence of ferroptosis will inevitably lead to lipid peroxidation.This article systematically summarizes the roles of iron death and oxidative stress in the development,progression,and treatment of multiple myeloma,with a view to providing new strategies for the treatment of multiple myeloma and overcoming drug resistance.
