核小体异常与淋巴瘤发生发展的分子机制
Molecular mechanisms of nucleosome abnormalities in the development and progression of lymphoma
【摘要】 表观遗传调控在血液系统恶性肿瘤的发生发展中起着重要作用。表观遗传学异常在淋巴瘤中很常见,包括核小体重塑、DNA甲基化和组蛋白修饰。在真核生物中,核小体是染色质的基本单位。染色质重塑复合物调节DNA可及性,并在基因表达调控中起重要作用。SWI/SNF复合物是真核生物重要的染色质重塑复合物,其亚基突变广泛存在于各种肿瘤中。染色质信号通路的失调是许多疾病和癌症的基础。文章总结了核小体异常在淋巴瘤发生发展的分子机制及其与肿瘤微环境形成之间的联系,以期为探索淋巴瘤发生发展中核小体的作用和靶向核小体的治疗方案提供理论依据。
【Abstract】 Epigenetic regulation plays a fundamental role in hematopoiesis and oncogenesis of hematologic malignancies. Epigenetic abnormalities are recurrent in lymphomas, including nucleosome remodeling, DNA methylation, and histone modifications. Nucleosomes, the basic units of chromatin in eukaryotes, regulate DNA accessibility and are implicated in gene expression. The SWI/SNF complex is a key chromatin remodeling complex in eukaryotes, and mutations in its subunits are frequently observed in various hematologic malignancies. Furthermore, dysregulation of chromatin signaling pathways forms the basis of many diseases and cancers. This review summarizes the molecular mechanisms of nucleosome abnormalities in lymphomagenesis and their connections with the tumor microenvironment, aiming to provide insights into the role of nucleosomes and potential therapeutic strategies targeting nucleosomes in the development of lymphomas.
