植物新品种‘赣龙1号’组培快繁技术研究
Study on the tissue culture and rapid propagation technology of new plant variety ‘Ganlong No.1’
【摘要】 【目的】为缓解樟属植物新品种‘赣龙1号’优质种苗短缺问题,建立一套高效的‘赣龙1号’组培快繁技术体系。【方法】以‘赣龙1号’嫩枝茎段为外植体,通过配制不同配比的培养基,开展腋芽诱导、愈伤组织诱导、丛生芽诱导及增殖、生根培养等研究,建立‘赣龙1号’组培快繁体系。【结果】(1)外植体适宜采集期为4―7月,无菌活体获得率高于50%。(2)腋芽诱导最适培养基为MS1+(2~3)mg/L 6-BA+(0.1~0.3)mg/L NAA,萌动率达67.6%。(3)愈伤组织诱导最佳培养基组合为MS1+2.0 mg/L 6-BA+0.2 mg/L NAA,诱导率为86.5%。(4)丛生芽诱导最佳培养基为MS1+0.4 mg/L6-BA+0.1 mg/L NAA,诱导率为84.2%。(5)丛生芽增殖阶段,培养基MS1+0.5mg/L 6-BA+0.1 mg/L NAA可使增殖系数达5.5。(6)生根培养以1/2 MS1+0.3 mg/L IAA为最佳,生根率达92.0%,平均根数2-3条/株。(7)组培苗在V泥炭土∶V黄心土为1∶2的基质中移栽成活率最高,达85.3%。【结论】以MS1为基本培养基,通过腋芽诱导—愈伤组织诱导—丛生芽诱导与增殖的途径,在1/2 MS1+0.3 mg/L IAA培养基上实现高效生根,并在泥炭土与黄心土(体积比1∶2)的混合基质中完成高成活率移栽,建立了‘赣龙1号’组培快繁体系。
【Abstract】 【Objective】To alleviate the shortage of high-quality seedlings of the new variety of Cinnamomum genus plant ‘Ganlong No. 1’, and establish an efficient tissue culture and rapid propagation technology system for ‘Ganlong No. 1’. 【Methods】Using tender stem segments of ‘Ganlong No.1’ as explants, experiments conducted on axillary bud induction, callus induction, clustered bud induction and proliferation, and rooting culture by formulating media with varying ratios, and established a rapid tissue culture propagation system for ‘Ganlong No.1’. 【Result】(1) The optimal period for explant collection was April to July, with a sterile viable explant yield exceeding 50%.(2) The most suitable medium for axillary bud induction was modified MS1 supplemented with 2-3 mg/L 6-BA and 0.1-0.3 mg/L NAA, achieving a budding rate of 67.6%.(3) The optimal combination for callus induction was MS1 + 2.0 mg/L 6-BA + 0.2 mg/L NAA, yielding an induction rate of 86.5%.(4) The optimal medium for inducing clustered buds was MS1+0.4 mg/L 6-BA+0.1 mg/L NAA, with an induction rate of 84.2%.(5) During the proliferation stage of clustered buds, MS1 + 0.5 mg/L 6-BA+ 0.1 mg/L NAA achieved a proliferation coefficient of 5.5.(6) For rooting culture, 1/2 MS1 + 0.3 mg/L IAA proved optimal, yielding a rooting rate of 92.0% with an average of 2-3 roots per plant.(7) Transplant survival rate was highest in tissue-cultured seedlings grown in a substrate of V(peat) ∶ V(yellowheart loam) = 1∶2, reaching 85.3%. 【Conclusion】Using MS1 as the basic medium, efficient rooting was achieved through axillary bud induction, callus induction, cluster bud induction, and proliferation on 1/2 MS1+0.3 mg/L IAA medium. High survival rate transplantation was completed in a mixed matrix of peat soil and yellow heart soil(1∶2), and the ‘Ganlong No.1’ tissue culture rapid propagation system was established.
