裙带菜中5种类胡萝卜素降解产物的抗炎活性研究(英文)

【摘要】 目的 鉴定裙带菜中的类胡萝卜素降解产物,评价其抗皮肤炎症活性。方法 利用多种色谱手段分离得到类胡萝卜素降解产物,运用NMR、MS等光谱学方法鉴定结构;通过建立脂多糖诱导的RAW264.7巨噬细胞、LPS-RAW264.7/CM诱导的HacaT细胞和HUVEC细胞炎症模型评价化合物的体外抗炎作用。结果 分离并鉴定5个类胡萝卜素降解产物,分别为3β-hydroxy-5α,6α-epoxy-β-ionone(1),(-)-loliolide(2),(+)-epiloliolide(3),grasshopper ketone(4)和apo-9’-fucoxanthinone(5)。化合物1~5能够不同程度抑制炎症状态下的RAW264.7细胞、HacaT细胞和HUVEC细胞释放NO、TNF-α和IL-6等炎症因子。结论 裙带菜中的类胡萝卜素降解产物具有抑制炎症细胞释放炎症因子、减轻炎症对表皮细胞和血管内皮细胞损伤的作用。

【Abstract】 Objective To identify the carotenoid degradation products in Undaria pinnatifida and evaluate their anti skin inflammatory activity. Methods Carotenoid degradation products were isolated by multiple chromatography methods and their structures were identified by spectroscopic methods such as nuclear magnetic resonance(NMR) and ESI-MS. The anti-inflammatory effect of these products was evaluated using lipopolysaccharide(LPS)-stimulated RAW264.7 cell model, LPS-RAW264.7/CM-induced HacaT or HUVEC cell model. Results Five compounds were isolated from U. pinnatifida and identified as 3β-hydroxy-5α,6α-epoxy-β-ionone(1),(-)-loliolide(2),(+)-epiloliolide(3), grasshopper ketone(4), and apo-9’-fucoxanthinone(5), respectively. Compounds 1-5 exerted different anti-inflammatory effects through reducing the levels of NO, TNF-α, and IL-6 in different cell models. Conclusion Five carotenoid degradation compounds from U. pinnatifida inhibited the release of inflammatory cytokines in inflammatory cells, and reduced the damage of inflammatory cytokines to epidermal cells and vascular endothelial cells.
