蒿柳花总多酚提取工艺及其体外抗氧化活性研究
Extraction process and in vitro antioxidant activity of total polyphenols from Salix viminalis
【摘要】 研究蒿柳花总多酚超声辅助提取工艺,并利用体外评价体系考察其抗氧化活性。在单因素实验基础上,利用Box-Behnken响应面设计模型对提取参数进行优化,获得最佳工艺;以VC为对照,进行DPPH·、ABTS+·清除实验来评价蒿柳花总多酚体外抗氧化活性。蒿柳花总多酚最佳的提取工艺为:乙醇浓度为50%超声时间55min,料液比1:35(g:mL),超声功率315W,在此工艺条件下得到的蒿柳花总多酚含量为3.05mg·g-1。所得蒿柳花总多酚对两种自由基均有较强的清除能力,当浓度为10μg·mL-1时,蒿柳花总多酚对DPPH·、ABTS+·的清除率分别为79.48%、101.47%,并高于同浓度VC的自由基清除能力。响应面法优化的蒿柳花多酚超声提取工艺稳定、简便易行,提取效率高,可指导其工业化生产。所得蒿柳花总多酚具有很强的抗氧化活性和潜在的开发利用价值。
【Abstract】 Study the ultrasound assisted extraction process of total polyphenols from Salix viminalis and evaluate their antioxidant activity using an in vitro evaluation system.On the basis of single-factor experiments,the Box Behnken design model was used to optimize the extraction parameters to obtain the optimal process;Using VC as a control,DPPH·and ABTS+·free radical scavenging tests were conducted to evaluate the antioxidant activity of total polyphenols from Salix viminalis.The optimal extraction process for total polyphenols from Salix viminalis flowers is:ethanol volume fraction of 50%,ultrasound time of 55 minutes,solid-liquid ratio of 1:35(g:mL),and ultrasound power of 315W.The total polyphenols content of Salix viminalis obtained by this process is 3.05mg·g-1.The total polyphenols from Salix viminalis have strong scavenging ability against two types of free radicals.When the concentration is 10μg·mL-1,the scavenging rates of Salix viminalis total polyphenols against DPPH·and ABTS+·free radicals are 79.48% and 101.47%,respectively,which is higher than the radical scavenging ability of VC at the same concentration.The ultrasonic extraction process of Salix viminalis polyphenols optimized by response surface methodology is stable,simple and feasible,with high extraction efficiency,which can guide its industrial production.The total polyphenols obtained from Salix viminalis have strong antioxidant activity and potential development and utilization value.
