高效液相色谱法测定道地产区金银花药材指纹图谱及化学模式识别分析
Determination of fingerprints and chemical pattern recognition of Flos Lonicerae in genuine areas by high performance liquid chromatography
【摘要】 建立了高效液相色谱测定道地产区金银花药材指纹图谱及化学模式识别分析方法。采用Phenomenex Synergi Hydro-RP80 A色谱柱(250 mm×4.6 mm,4μm),柱温为25°C,梯度洗脱以流量为1.0 mL/min的乙腈-0.1%磷酸溶液为流动相,检测波长为327nm,建立25批不同产地金银花药材的指纹图谱,并用聚类分析和主成分分析等统计学研究方法以及《中药色谱指纹图谱相似度评价软件系统》(2012年版)对25批金银花样品进行质量分析,标示了13个共有峰,指认了其中的6个共有峰,25批金银花样品中有88%的样品相似度大于0.91。对25批三大道地产区的金银花样品进行的聚类分析和主成分分析,结果显示A与B产区的金银花药材的成分均一性总体低于C产区的金银花药材,同时研究了金银花与山银花的混淆区分,为金银花的掺伪鉴别提供参考。该方法可对金银花药材内在质量做出评价,进一步加强了金银花药材的质量控制,为药材的鉴别、品质评价、市场规范、中药制剂安全性、行业标准化等方面提供重要的理论基础。
【Abstract】 The fingerprint and chemical pattern recognition analysis method of Flos Lonicerae in genuine areas were established by high performance liquid chromatography. The Phenomenex Synergi Hydro-RP80A chromatographic column(250 mm ×4.6 mm, 4 μm) was used, with the column temperature of 25° C. Gradient elution was carried out using acetonitrile-0.1% phosphoric acid solution with the flow rate of 1.0 m L/min as the mobile phase, and the detection wavelength was 327nm. The fingerprints of 25 batches of Flos Lonicerae from different origins were established, and their quality were analyzed by using statistical methods such as cluster analysis and principal component analysis and the software system for similarity evaluation of chromatographic fingerprints of traditional Chinese medicine(2012 version). 13 common peaks were identified, and 6 of which were identified, the similarity of 88% of the 25 batches of Flos Lonicerae samples were more than 0.91. Cluster analysis and principal component analysis were carried out on 25 batches of honeysuckle samples from 3genuine producing areas. The results showed that the composition uniformity of honeysuckle of A and B was lower than that of C. At the same time, the confusion between honeysuckle and honeysuckle was studied, which provided reference for the adulteration identification of honeysuckle. This method can evaluate the internal quality of Flos Lonicerae, further strengthen the quality control of Flos Lonicerae, and provide an important theoretical basis for the identification, quality evaluation,market regulation, safety of traditional Chinese medicine preparations, and industry standardization of Flos Lonicerae.
