红叶石楠组培快繁体系的建立

【摘要】 选取衡水金树园林绿化科技有限公司苗圃场生长健壮的红叶石楠新生腋芽为外植体,探讨不同消毒处理方式、不同培养基配方、不同炼苗移栽条件对红叶石楠组培快繁体系的影响,筛选出红叶石楠外植体的最佳消毒剂、消毒时间及不定芽分化培养基配方,以及红叶石楠组培苗的最佳生根培养基配方、栽培基质组合,为红叶石楠规模化育苗提供一定的技术支持。研究结果表明:0.1%HgCl2消毒7 min时,红叶石楠外植体接种成活率最高,为57.8%;不定芽分化最佳培养基配方为MS+2.0 mg/L 6-BA+0.2mg/L NAA+0.4 mg/L KT,不定芽分化诱导率高达83%;红叶石楠组培苗最佳生根培养基配方为1/2 MS+0.1 mg/L NAA+0.5 mg/L IBA,生根率为96%,平均根长为7.66 cm;红叶石楠组培苗最佳栽培基质组合为园土∶腐叶土∶珍珠岩=5∶4∶1,成活率高达92%。

【Abstract】 The strong new axillary buds of healthy red-leaf photinia grown in the nursery of Hengshui Jinshu Landscape Greening Technology Co,Ltd.were selected as explants,and explore the effects of different disinfection methods,different medium formulations,and different acclimatization and transplantation conditions on the tissue culture fast multiplication system of red-leaf photinia.The study aims to identify the optimal disinfectant,disinfection time,and adventitious bud differentiation medium formulation for red-leaf photinia explants,as well as the optimal rooting medium formulation and substrate combination for red-leaf photinia tissue culture seedlings,providing technical support for the large-scale cultivation of red-leaf photinia.The research results are as follows:0.1%HgCl2 treatment for 7 minutes has the best disinfection effect on red-leaf photinia explants,with a low pollution rate of 13.3%and the highest survival rate of 57.8%.The optimal medium for adventitious bud differentiation is MS+2.0mg/L 6-BA+0.2mg/L NAA+0.4mg/L KT,with an adventitious bud induction rate of up to 83%.The most suitable rooting medium is 1/2 MS+0.1mg/L NAA+0.5mg/L IBA,with a rooting rate of 96%and an average root length of 7.66cm.The best transplanting substrate combination is garden soil:leaf mold:perlite=5:4:1,with a high survival rate of seedlings up to 92%.A fast multiplication technology system for red-leaf photinia tissue culture seedlings has been established preliminarily,providing a theoretical basis for achieving large-scale production.
