湖南省玉竹叶斑病病原菌的分离鉴定

【摘要】 为明确湖南省玉竹叶斑病的病原菌,研究以当地玉竹病叶为试验材料,采用组织分离法分离纯化菌株,采用柯赫氏法则(Koch’s postulates)验证其致病性,通过菌落形态观察、孢子和菌丝显微特征分析,结合分子生物学鉴定方法对该菌株进行鉴定。结果显示:分离得到的菌株YZy形态特征与木贼镰刀菌(Fusarium equiseti)一致,菌落正面呈米白色,气生菌丝呈棉絮状,菌落背面呈淡黄色,大分生孢子呈纺锤形或镰刀形,小分生孢子数量少,厚垣孢子呈球形且表面粗糙;致病性测定与田间症状相符;通过ITS、EF-1α和RPB1多位点系统发育分析发现,该菌株与木贼镰刀菌(MH11)聚在同一分支,同源性达87%。综上,湖南省玉竹产区叶斑病的致病菌为木贼镰刀菌,这一发现为该病害的有效防治提供了参考。

【Abstract】 This study aims to identify the pathogen causing leaf spot in Polygonatum odoratum in Hunan Province. The pathogen was isolated from the diseased leaves of P. odoratum by the tissue culture method, and its pathogenicity was verified according to Koch’s postulates. The isolate was identified by morphological observation of colonies, conidia, and hyphae and molecular biological methods.The results showed that the obtained strain YZy had the morphological characteristics consistent with those of Fusarium equiseti.Specifically, the strain presented colonies creamy white on the front and light yellow at the back, flocculent aerial hyphae, spindleshaped or sickle-shaped macroconidia, few microconidia, and sphere chlamydospores with rough surfaces. The symptoms caused by the strain in laboratory were consistent with those in the field. The multilocus phylogenetic analysis based on ITS, EF-1α, and RPB1showed that the strain shared the same clade(87% homology) with F. equiseti. Therefore, the pathogen causing leaf spot in P. odoratum is F. equiseti, which provides reference for the effective control of this disease.
