羧甲基茯苓多糖的分离纯化、结构表征及其抗氧化作用

【摘要】 为探究羧甲基茯苓多糖(Carboxymethyl Pachymaran,CMP)的结构表征及其抗氧化作用,以CMP为试验材料,并对其分离纯化后的组分进行结构表征和抗氧化作用检测。结果表明,CMP经二乙氨基乙基(diethylaminoethyl,DEAE)纤维素离子交换层析柱纯化后得到两种组分CMP0和CMP5。将主要成分CMP5用葡聚糖凝胶G-100层析柱纯化,结合紫外分光光度测定法、傅里叶红外光谱法和高效凝胶渗透色谱法等方法对其结构表征进行检测分析。结果表明,CMP5的多糖含量为92.07%,分子量为75.724 kDa,红外光谱检测出糖类特征峰与羧甲基基团特征峰,单糖组成分析表明CMP5是一种葡聚糖。以CMP5的DPPH清除率、羟基自由基的清除能力、总还原能力和Fe2+螯合能力作为其体外抗氧化作用的检测指标,检测结果显示,CMP5对DPPH自由基清除率为58.25%,对羟基自由基清除率为55.41%,对Fe2+螯合率为39.89%。综上所述,CMP5 具有较好的抗氧化作用,是一种潜在的抗氧化剂。

【Abstract】 This study aims to investigate the structural characteristics and antioxidant activity of carboxymethyl pachymaran (CMP).With CMP as the test material,the isolated and purified components were detected for their structural characteristics and antioxidant activity.CMP was purified by diethylaminoethyl (DEAE) cellulose ion-exchange chromatography to give two components,CMP0 and CMP5.Subsequently,CMP5 was purified by dextran gel G-100 chromatography,and the structural characterization was conducted by UV spectrophotometry,Fourier infrared spectroscopy,and high-performance gel permeation chromatography.The results showed that the polysaccharide content and molecular weight of CMP5 were 92.07% and 75.724 kDa,respectively.The characteristic peaks of sugar and carboxymethyl group were detected by infrared spectroscopy,and the monosaccharide composition analysis suggested that CMP5 was a dextran.The DPPH free radical scavenging rate,hydroxyl radical scavenging rate,total reducing capacity,and Fe2+ chelating capacity were used as indicators for assessing the antioxidant activity of CMP5 in vitro.The results showed that CMP5 had the DPPH free radical scavenging rate,hydroxyl radical scavenging rate,and Fe2+ chelating rate of 58.25%,55.41%,and 39.89%,respectively.In summary,CMP5 has good antioxidant activity and is a potential antioxidant.
