织金白鹅的基因组调研分析

【摘要】 为了研究织金白鹅的起源、进化及适应性,确定适合该物种的全基因组测序和组装策略,通过低覆盖度基因组Survey测序,评估织金白鹅基因组的大小及杂合度,以确定适合织金白鹅全基因组的测序策略,试验采用第二代高通量测序技术(Illumina HiseqTM 2000),对织金白鹅基因组大小、杂合度、重复序列、GC含量等基因组信息进行了检测。结果表明:测序获得了66.16 Gb织金白鹅基因组测序数据,基因组大小约为1.11 Gb,测序深度为59.57X,GC含量为42.08%;从K-mer分布曲线发现织金白鹅基因组有明显的杂合峰,杂合度达0.62%,重复序列含量约为11.44%;对织金白鹅的质体进行评估,发现核外DNA含量很低。说明研究结果为揭示织金白鹅的起源、进化及基本信息具有重要意义,且为织金白鹅物种进化、倍性水平和全基因组测序等研究提供理论依据。

【Abstract】 In order to study the origin, evolution and adaptability of Zhijin white geese to determine the whole genome sequencing and assembly strategy suitable for this species, and evaluate Zhijin white goose genome size and heterozygosity through low coverage genome Survey sequencing to determine the suitable whole genome sequencing strategy for Zhijin white goose, in this study, the genomic information such as genome size, heterozygosity, repeat sequence and GC content of Zhijin white geese was studied by Illumina HiseqTM 2000(the second generation of high-throughput sequencing). The results showed that 66.16 Gb genome sequencing data of Zhijin white geese was obtained; the estimated genome size was about 1.11 Gb; the sequencing depth was 59.57X, and GC content was 42.08%. From the K-mer distribution curve, it was found that there was an obvious heterozygosity peak in the genome of Zhijin white geese, with a heterozygosity rate of 0.62% and a repeat sequence content of 11.44%. The plastid of Zhijin white goose was evaluated, and it was found that the extracellular DNA content was very low. The results of this study were of great significance to reveal the origin, evolution and essential information of Zhijin white geese, and provide a theoretical basis for the follow-up study of species evolution, ploidy level and whole genone sequencing of Zhijin White geese.
