新型色谱柱技术对液相色谱基础实验的提升
New Column Technology and Its Developments in Basic Experiment of Liquid Chromatography
【摘要】 在本科生仪器分析实验教学中,受限于仪器效率与教学时长,许多教学内容无法展开,合理安排并丰富教学内容是教学领域的难点。例如每个单样的色谱分离时间长,是本科生液相色谱基础实验的瓶颈。通过自研的快速传质色谱柱,一个样品在3 min内可完全分离。实践不明:新型色谱柱技术的采用,显著提升了仪器效率,在教学时长不变的情况下,教学内容获得了极大的拓展:30 min即可快速完成实验样品的定性分析和色谱柱的基本性能评价;进而把优化色谱条件从验证型改为研究型实验;最后用外标工作曲线完成样品的定量分析。改进后实验教学内容充实深入,知识点覆盖面广,增强了基本操作实践,培养了科研思维,为液相色谱的实际应用打下良好基础。相关实验的设计为提升仪器分析实验教学效果提供了示范。
【Abstract】 In basic Instrumental Analysis experiments for undergraduate students, many contents of courses can’t be carried out because of restriction in instrument efficiency and teaching duration. For instance, in HPLC instrument, the separation of samples is a time-consuming process. As a result, it cannot arrange many tasks in an experiment. By the homemade column with fast mass transfer property, a sample can be separated entirely in 3 min. The new column technology increased the instrument efficiency obviously, then the teaching content can be expanded sufficiently. The qualitative analysis of experimental samples and the evaluation of chromatographic column was achieved quickly in 30 min. Therefore, this had a chance to optimize chromatographic conditions from verification to inquiry experiment. Finally, the results of the quantitative analysis were carried out by the working curve method. According to the improvement, the fundamental experimental content was enriched and deepened, the hands-on practice of basic operations was enhanced. It can lay a solid foundation for the practical application of liquid chromatography. It is an example to promote the effect of instrumental analysis experiments.
