三羟甲基丙烷体外细胞毒性及致突变性试验研究

【摘要】 目的 该研究旨在探讨分析三羟甲基丙烷(trimethylolpropane,TMP)的体外细胞毒性和致突变性,为安全使用TMP提供毒理学数据。方法 通过观察细胞形态、存活率及细胞内活性氧(reactive oxygen species,ROS)的变化,判断TMP对中国仓鼠肺细胞(chinese hamster lung cells,CHL)的毒性。通过细菌回复突变试验(bacterial reverse mutation test,Ames test)和双腔室玻片系统的体外染色体畸变试验,观察其体外致突变性。结果 随着TMP染毒剂量增加,镜下观察到贴壁细胞逐渐固缩变圆或脱落,细胞数量逐渐减少。通过细胞存活率计算出TMP对CHL细胞半数抑制浓度(semi-inhibitory concentration,IC50)为16.1 mg/mL(+S9,6 h)和14.2 mg/mL(-S9,24 h)。ROS试验发现,当TMP的质量浓度≥5 mg/mL时,可诱导CHL细胞产生ROS。Ames试验结果提示,在±S9条件下,各剂量组对TA97a、TA98、TA100、TA102四株试验菌株均无致突变作用。体外染色体畸变试验显示,在±S9(6 h)和-S9(24 h)三种处理方式下,各剂量组染色体畸变率均小于5%,与本组溶剂对照组染色体畸变率比较,差异无统计学意义(χ2=0、0、0.128、0、1.523、0.253、0、0、1.375,P>0.05)。结论 在该试验条件下,TMP在高浓度可使贴壁细胞形态发生改变或死亡,并能使细胞内ROS含量增加,但无明显致突变作用。

【Abstract】 Objective To study the cytotoxicity and mutagenicity of trimethylolpropane(TMP)in vitro,and to provide toxicological data for its safety use.Methods The morphological changes of cells and reactive oxygen species(ROS)were observed,and the survival rate was calculated by MTT test.Ames test and two well chamber slides-chromosome aberration test were used to detect the mutagenicity of TMP.Results After exposure to different concentrations of TMP,with the increase of concentration,the cell morphology gradually became round until most of the cells were autolysed.The semi-inhibitory concentration(IC50)tested by MTT were 16.1 mg/mL(+S9,6 h)and 14.2 mg/mL(-S9,24 h). When the concentration of TMP is greater than or equal to 5 mg/mL,it can induce CHL cells to produce ROS.Ames test indicated that each dose group had no mutagenic effect on the four test strains(TA979a,TA98,TA100,TA102)with or without the metabolic activation system(S9).The results of the in vitro chromosome aberration test showed that under the three treatment methods of ±S9(6 h)and-S9(24 h),there were no statistical significance in chromosome aberration rates of CHL cells compared with negative control group(χ2=0,0,0.128,0,1.523,0.253,0,0,1.375,P>0.05).Conclusion Under the conditions of this experiment,morphological changes of cells and an increase of ROS were observed at high concentrations, but no evidence of mutagenicity were found.
