金花茶花朵总黄酮的提取工艺优化及抗氧化、抗炎活性研究

【摘要】 旨在高效开发利用金花茶花朵资源,优化其总黄酮的提取工艺,并系统评价其抗氧化及抗炎活性。研究以总黄酮含量为评价指标,采用单因素及响应面试验优化回流提取法提取金花茶花朵总黄酮的工艺;通过测定总黄酮提取物对·OH、DPPH·、ABTS~+·的清除效果,评价其抗氧化活性;并分析金花茶花朵总黄酮提取物对脂多糖诱导的小鼠单核巨噬细胞白血病细胞(RAW 264.7)一氧化氮(NO)生成的抑制效果以及对TNF-α、IL-1β和IL-6水平的影响。结果表明,最优提取工艺参数为:乙醇体积分数65%,料液比1∶35 g/mL,提取时间8 h;在此条件下,从金花茶花朵中提取的总黄酮含量达11.39 mg/g。活性研究结果表明,金花茶总黄酮对3种自由基均呈现浓度依赖性的清除作用,并显著抑制脂多糖诱导的RAW 264.7巨噬细胞NO生成,同时抑制促炎因子TNF-α、IL-1β和IL-6的分泌。本研究为金花茶花朵在功能性食品和药品领域的深入开发提供了理论依据。

【Abstract】 The study aims to efficiently develop and utilize the flower resources of Camellia nitidissima, optimize the extraction process of its total flavonoids, and systematically evaluate its antioxidant and anti-inflammatory activities. In this study, the extraction conditions of total flavonoids from Camellia nitidissima flowers were optimized using a reflux extraction method based on single-factor experiments and response surface methodology, with the total flavonoid content as the evaluation index. The antioxidant activity was assessed by determining the scavenging effects of the flavonoid extract on ·OH, DPPH·, and ABTS~+· radicals. In addition, the anti-inflammatory activity was evaluated by analyzing the inhibitory effect of the extract on nitric oxide(NO) production and the levels of TNF-α, IL-1β, and IL-6 in LPS-induced RAW264.7 macrophages. The results showed that the optimal extraction parameters were as follows: ethanol concentration of 65%, solid-to-liquid ratio of 1∶35 g/mL, and reflux extraction time of 8 h. Under these conditions, the total flavonoid content reached 11.39 mg/g. The bioactivity assays indicated that the total flavonoids from Camellia nitidissima flowers exhibited dose-dependent scavenging effects on all three radicals, significantly inhibited NO production in LPS-induced RAW264.7 cells, and suppressed the secretion of pro-inflammatory cytokines TNF-α, IL-1β, and IL-6. This research provides a theoretical basis for the in-depth development of Camellia nitidissima flowers in the fields of functional foods and medicines.
