黄连解毒汤对热体变应性鼻炎大鼠骨髓干细胞增殖的影响
Effects of Huanglian Detoxification Decoction on Proliferation of Bone Marrow Stem Cells in Heat Constitutional Type Rats with Allergic Rhinitis
【摘要】 目的:研究黄连解毒汤对热体变应性鼻炎大鼠骨髓间充质干细胞(bone marrow mesenchymal stem cells,BMSCs)和骨髓内皮祖细胞(bone endothelial progenitor cells,BEPCs)增殖的影响。方法:复制变应性鼻炎的遗传性热体大鼠模型,将48例模型大鼠分为热体无干预组和热体干预组,雌雄各半,每组24例。热体干预组于建模次日开始予黄连解毒汤灌胃,连续干预7天;热体无干预组不做灌胃处理。在干预的第0、1、3、7天评价两组大鼠鼻部症状评分,并取材检测BMSCs和BEPCs的含量。结果:干预后与同组第0天比较,热体干预组在第3天及第7天,鼻部症状评分减少(P<0.05);与同一时间节点热体无干预组比较,热体干预组大鼠在第7天时,鼻部症状评分减少(P<0.05);与同组第0天比较,热体无干预组大鼠BMSCs与BEPCs在第1、3、7天减少(P<0.05),热体干预组大鼠BMSCs在第7天增多(P<0.05),BEPCs在第1天减少(P<0.05);与同一时间节点热体无干预组比较,热体无干预组大鼠第3、7天BMSCs与BEPCs增多(P<0.05)。结论:黄连解毒汤能够促进热体大鼠BMSCs、BEPCs增殖表达,有效缓解变应性鼻炎的症状。
【Abstract】 Objective: To study the effects of Huanglian detoxification decoction on the proliferation of bone marrow mesenchymal stem cells(BMSCs) and bone endothelial progenitor cells(BEPCs) in heat constitutional type rats with allergic rhinitis(AR). Methods: After depulicating hereditary heat constitutional type rats with AR, 48 rat models were allocated to heat constitution group without intervention and heat constitution group with intervention, half of male and half of female, 24 cases in each group. Heat constitution group with intervention was drenched with Huanglian detoxification decoction since the second day of modeling, for seven days continuously; heat constitution group without intervention remained unhandled. The scores of nasal symptoms of the two groups were evaluated on the zero, first, third and seventh day of intervention, and the contents of BMSCs and BEPCs were detected after taking the tissues. Results: After the intervention, compared with the same group on the zero day, the scores of nasal symptoms in heat constitution group with intervention were reduced on the third and seventh day(P<0.05); compared with heat constitution group without intervention at the same time point, the reduction in the scores of nasal symptoms in heat constitution group with intervention was significant on the seventh day(P<0.05); compared with the same group on the zero day, the decrease in the expressions of BMSCs and BEPCs was evident in heat constitution group without intervention on the first, third and seventh day(P<0.05), the increase in the expressions of BMSCs was obvious in heat constitution group with intervention on the seventh day(P<0.05), while the expressions of BEPCs were lowered(P<0.05). The comparison with constitution group without intervention at the same time point showed that the levels of BMSCs and BEPCs were elevated in heat constitution group without intervention on the third and seventh day(P<0.05). Conclusion: Huanglian detoxification decoction could promote the proliferation of BMSCs and BEPCs, and effectively alleviate the symptoms of AR.
