基于方剂反证策略的多囊卵巢综合征肾虚血瘀证大鼠模型构建与评价
Construction and evaluation of polycystic ovary syndrome rat model with kidney deficiency and blood stasis based on prescription counter-evidence strategy
【摘要】 目的 建立多囊卵巢综合征(PCOS)肾虚血瘀证病证结合动物模型,并对其进行评价。方法 将SD大鼠随机分为正常组、PCOS组、模型组和方剂反证组,每组10只。正常组每日灌胃给予1 mg·kg-1的1%羧甲基纤维素钠水溶液;PCOS组每日灌胃给予1 mg·kg-1的来曲唑;模型组在PCOS组的基础上,自第21天起每日皮下注射0.6 mg·kg-1的盐酸肾上腺素溶液,连续注射2周;方剂反证组在模型组的基础上,自第21天起每日灌胃给予12.51 g·kg-1的朱氏调经汤2周。干预35 d后,比较各组大鼠的卵巢指数、睾酮、维生素D、全血黏度低切值等指标。结果 正常组、PCOS组、模型组和方剂反证组的卵巢指数分别为(6.39±0.58)×10-4、(5.32±0.71)×10-4、(5.05±0.29)×10-4和(8.17±0.65)×10-4,睾酮水平分别为(7.10±0.97)、(9.22±0.43)、(9.06±1.21)和(7.73±0.76)nmol·L-1,维生素D水平分别为(1 850.98±227.95)、(986.10±152.50)、(1 220.67±64.35)和(1 598.20±294.75)pg·mL-1,全血黏度低切值分别为(13.00±1.79)、(13.37±0.54)、(16.38±1.51)和(11.43±2.77)mPa·s。模型组的上述指标与方剂反证组和正常组比较,在统计学上差异均有统计学意义(均P<0.05)。结论 用来曲唑联合盐酸肾上腺素的方式可以稳定复制具有肾虚血瘀证候特征的PCOS大鼠模型。
【Abstract】 Objective To establish an animal model of polycystic ovary syndrome (PCOS) characterized by kidney deficiency and blood stasis,and to evaluate its efficacy.Methods SD rats were randomly divided into normal group,PCOS group,model group and formula counterevidence group,with 10 rats in each group.The normal group was given1 mg·kg-11%sodium carboxymethyl cellulose daily by intragastric administration;PCOS group was given 1 mg·kg-1letrozole daily by intragastric administration;on the basis of PCOS group,the model group was injected subcutaneously with 0.6 mg·kg-1epinephrine hydrochloride solution daily for 2 weeks from day 21.On the basis of the model group,the formula counter-evidence group was given 12.51g·kg-1Zhu’s Tiaojing decoction daily for 2 weeks from the 21st day.After 35 days of intervention,the indexes of ovarian index,testosterone,vitamin D and whole blood low shear rate were compared.Results The ovarian indexes of normal,PCOS,model and formula counter-evidence groups were (6.39±0.58)×10-4,(5.32±0.71)×10-4,(5.05±0.29)×10-4and (8.17±0.65)×10-4;testosterone concentrations were(7.10±0.97),(9.22±0.43),(9.06±1.21) and (7.73±0.76) nmol·L-1;vitamin D levels were(1 850.98±227.95),(986.10±152.50),(1 220.67±64.35) and (1 598.20±294.75) pg·m L-1;whole blood low shear rates were (13.00±1.79),(13.37±0.54),(16.38±1.51) and (11.43±2.77) m Pa·s,respectively.The above indexes in the model group were statistically significant compared with the formula counter-evidence group and the normal group (all P<0.05).Conclusion Letrozole combined with epinephrine hydrochloride can stably replicate the PCOS rat model with kidney deficiency and blood stasis syndrome.
