2个乌鸦果居群组培快繁的比较研究

【摘要】 为了掌握乌鸦果组培快繁技术,促进我国乌鸦果野生种质资源保存和合理开发利用,对云南省丽江市古城区贵峰社区和玉龙县鲁甸乡2个乌鸦果居群(分别简称贵峰、鲁甸居群)进行组织培养和快速繁殖技术研究。以居群土壤pH值分别为5.90、7.68的2个乌鸦果居群幼嫩枝条茎段作为外植体,在pH值5.5添加不同植物生长调节剂的威斯特摩尔培养基(WPM)上进行初代培养、继代培养及瓶内生根培养,采用不同生根剂及基质进行瓶外生根。结果表明,供试2个乌鸦果在pH值5.5的培养基中均能较好地诱导分化及增殖,只加玉米素(ZT)而不添加其他植物生长调节剂更有利于乌鸦果诱导分化及增殖,WPM+1.0 mg/L ZT是2个乌鸦果居群初代培养较佳的诱导培养基,WPM+0.5 mg/L ZT是其较佳的增殖培养基。2个乌鸦果居群瓶内生根培养差异较大,不同IBA浓度对2个乌鸦果居群瓶内生根率影响差异显著,居群土壤pH值7.68的贵峰乌鸦果试管苗在pH值5.5的培养基中难生根,而居群土壤pH值5.90的鲁甸乌鸦果试管苗可正常生根,1/2WPM+0.02 mg/L IBA是其较佳的生根培养基。2个乌鸦果居群瓶外生根时均能较好生根,两者差异较小,进口苔藓及3-吲哚丁酸钾(IBA-K)较有利于其瓶外生根。

【Abstract】 To master the tissue culture and rapid propagation techniques of Vaccinium fragile Franch. and promote the conservation and rational development and utilization of its wild germplasm resources in China, a study was conducted on the tissue culture and micropropagation technology of two V. fragile Franch. populations in Guifeng community of Gucheng District of Lijiang City, and Ludian Village in Yulong County. Young stem segments from two V. fragile Franch. populations with the soil pH values of 5.90 and 7.68, respectively, were used as explants. Primary culture, subculture and in vitro rooting were conducted on a WPM(Woody Plant Medium) medium with a pH value of 5.5 and supplemented with different plant growth regulators. Additionally, different rooting agents and substrates were applied for ex vitro rooting. The results showed that both V. fragile Franch. populations exhibited satisfactory differentiation and proliferation on the culture medium with pH 5.5. The medium supplemented solely with zeatin(ZT) was more favorable for inducing differentiation and proliferation of V. fragile Franch.. The optimal induction medium for initial culture of both V. fragile Franch. populations was WPM supplemented with 1.0 mg/L ZT, with WPM + 0.5 mg/L ZT being the optimal proliferation medium. Significant differences were observed in in vitro rooting between two populations. The effect of different IBA concentrations on the in vitro rooting rate showed significant differences between the two V. fragile Franch. populations. The plantlets of V. fragile Franch. from Guifeng with soil pH 7.68 showed extremely poor rooting on a rooting medium with pH 5.5. Plantlets of V. fragile Franch. from Ludian with soil pH 5.9 could normally root on that rooting medium, and 1/2 WPM + 0.02 mg/L IBA was the better rooting medium. The plantlets of both populations exhibited good rooting performance with little difference with ex vitro rooting methods. The imported moss and IBA-K were better for ex vitro rooting of both V. fragile Franch. populations.
