杧果MiCLH1基因的时空表达模式分析
Temporal and spatial expression analysis of the MiCLH1 gene in mango
【摘要】 叶绿素酶(CLH)是叶绿素分解代谢途径中的关键酶。本研究以“台农一号”杧Mangifera indica L.为材料,分析MiCLH1基因结构、编码蛋白理化性质、跨膜结构域、信号肽、磷酸化位点,系统进化、顺式作用元件等,测定采后1μL/L 1-甲基环丙烯和5 g/L乙烯处理的杧果果皮叶绿素、类胡萝卜素含量,叶绿素酶活性,及MiCLH1基因表达量。结果表明,MiCLH1全长960 bp,编码氨基酸320个,等电点为5.62,蛋白质分子量为34.08 kD。序列比对和进化树分析表明,MiCLH1与阿月浑子同源性最高。启动子序列分析显示,MiCLH1基因的启动子序列中激素响应元件含有乙烯响应元件。1-甲基环丙烯处理能够延缓采后果实果皮叶绿素降解,抑制类胡萝卜素合成和MiCLH1表达,乙烯处理能够促进采后果实果皮叶绿素降解、类胡萝卜素合成和MiCLH1表达,初步推断MiCLH1可能参与调控杧果果实脱绿转色过程。
【Abstract】 Chlorophyllase(CLH) is a key enzyme in chlorophyll catabolism pathway. In this study, Tainong No. 1 mango was used as material to analyze the gene structure of MiCLH1, including physicochemical properties of its encoded protein, transmembrane domains, signal peptides, phosphorylation sites, phylogenetic evolution, and cis-acting element, and determine the MiCLH1 gene expression levels, the chlorophyll and carotenoid content, chlorophyllase activity of fruit peels treated with 1 μL/L 1-methylcyclopropene(1-MCP) and 5 g/L ethephon. The results showed that MiCLH1 was 960 bp in length, encoded 320 amino acids, with an isoelectric point of 5.62, and its protein molecular weight was 34.08 kD. Sequence alignment and evolutionary tree analysis showed that MiCLH1 had the highest homology with its homologous in pistachio. Promoter sequence analysis showed that hormone response elements in the promoter of MiCLH1 gene included ethylene response elements. 1-MCP treatment could delay chlorophyll degradation in postharvest fruit peel, inhibit carotenoid synthesis and MiCLH1 expression. In contrast, ethylene treatment accelerated the chlorophyll degradation in fruit peel, promoted carotenoid synthesis and MiCLH1 expression. These results suggested that MiCLH1 may play a regulatory role in the degreening and color transition process of mango fruit.
