代谢工程改造酵母合成单萜类化合物研究进展
Research Progress on Metabolic Engineering of Yeast for Monoterpenoids Synthesis
【摘要】 单萜是一种由两个异戊二烯单位组成分子骨架的化合物,一般广泛存在于高等植物的分泌组织。其含氧衍生物如醇类、醛类、酮类具有很高的生物活性,常被用于制造防腐剂、除味剂、消毒剂等。单萜的获取通常依赖于天然产物提取和化工合成这两种传统方式,但随着合成生物学技术的不断革新与发展,利用微生物作为代谢工厂进行单萜生产已是大势所趋,具有成本低、可控性高、上升空间大等优点。从代谢途径工程、酶工程、细胞工程3个方面概述了以酵母为底盘生产单萜类化合物的研究进展,展示了代谢工程改造手段对于天然活性产物在微生物生产中的普适性和有效性。
【Abstract】 Monoterpenoids are a class of compounds with a molecular structure consisting of two isoprene units, containing a total of 10 carbon atoms.They are commonly found in the secretory tissues of higher plants.Their oxygenated derivatives, such as alcohols, aldehydes, and ketones, exhibit high biological activity and are frequently used to produce preservatives, deodorants, disinfectants, and other products.Traditionally, monoterpenoids have been obtained through natural product extraction and chemical synthesis.However, with the continuous innovation and development of synthetic biology technology, the use of microorganisms as metabolic factories for production has become an inevitable trend, offering advantages such as low cost, high controllability, and significant potential for scalability.This review summarizes the research progress in the production of monoterpenoids using yeast as a chassis, focusing on three aspects: metabolic pathway engineering, enzyme engineering, and cell engineering, demonstrating the applicability and effectiveness of metabolic engineering transformation methods for the production of natural active products in microorganisms.
