紫玉米花青素对赤水乌骨鸡盲肠短链脂肪酸与微生物组成的影响
Effects of Purple Corn Anthocyanin on Cecum Short-Chain Fatty Acids and Microbial Composition in Chishui Black-Bone Chicken
【摘要】 试验旨在研究紫玉米花青素(purple corn anthocyanin, PCA)对赤水乌骨鸡盲肠短链脂肪酸(short-chain fatty acids,SCFAs)、菌群多样性、微生物组成和丰富度的影响。采用单因素完全随机试验设计,选取144只体况良好、体重相近的8周龄赤水乌骨鸡,随机分为2组,对照组饲喂基础饲粮,试验组在基础饲粮中添加80 mg/kg的PCA,每组6个重复,每个重复12只鸡。试验持续75 d,其中预试期15 d,正试期60 d。结果表明:(1)添加80 mg/kg PCA显著提高了盲肠中丙酸、异丁酸、丁酸、异戊酸、戊酸和总短链脂肪酸含量(P<0.05)。(2)在属水平上,拟杆菌属为优势菌属,添加80 mg/kg PCA显著提高了拟杆菌属、副拟杆菌属和乳杆菌属等有益菌群的相对丰度(P<0.05)。(3)在门水平上,WPS-2菌门相对丰度与异戊酸、丙酸、异丁酸含量存在显著负相关(P<0.05),弯曲杆菌门(Campilobacterota)相对丰度与异己酸含量存在显著正相关(P<0.05);在属水平上,unclassified_o_Bacteroidales菌属相对丰度与异己酸含量存在显著负相关(P<0.05)。由此可见,饲粮中添加PCA可显著改善盲肠内容物中短链脂肪酸含量与组成,显著提高拟杆菌属、副拟杆菌属和乳杆菌属菌群的相对丰度。
【Abstract】 The experiment aimed to investigate the effects of purple corn anthocyanin(PCA) on shortchain fatty acids(SCFAs), microbiota diversity, microbial composition, and richness in cecum in Chishui black-Bone chickens. A single-factor completely randomized trial design was adopted. A total of 144 healthy, similarly weighted 8-weekold Chishui Black-Bone Chickens were randomly divided into two groups: the control group was fed a basal diet, while the experimental group was fed a basal diet supplemented with 80 mg/kg of purple corn anthocyanin. Each group had six replicates, with 12 chickens per replicate. The experiment lasted for 75 days, with a pre-feeding period of 15 days and a trial period of 60 days. The results showed that:(1) adding 80 mg/kg PCA significantly increased the levels of propionic acid, isobutyric acid, butyric acid, isovaleric acid, valeric acid, and total short-chain fatty acids in the cecum(P<0.05).(2) At the genus level, Bacteroides was the dominant genus, and the addition of 80 mg/kg PCA significantly increased the relative abundance of beneficial bacterial groups such as Bacteroides, Parabacteroides, and Lactobacillus(P<0.05).(3) At the phylum level, the relative abundance of the WPS-2 phylum was significantly negatively correlated with the levels of isovaleric acid, propionic acid, and isobutyric acid(P<0.05); the relative abundance of the Campilobacterota phylum was significantly positively correlated with the level of isocaproic acid(P<0.05). At the genus level, the relative abundance of the unclassified_o_Bacteroidales genus was significantly negatively correlated with the level of isocaproic acid(P<0.05). These findings indicate that supplementing the diet with purple corn anthocyanins significantly improved the content and composition of short-chain fatty acids in cecum contents and significantly increased the relative abundance of Bacteroides, Parabacteroides, and Lactobacillus.
