基于网络药理学探讨三虫半夏白术天麻汤治疗缺血性脑血管病的作用机制

【摘要】 目的 采用网络药理学探讨三虫半夏白术天麻汤治疗缺血性脑血管病(ICVD)的作用机制。方法 结合TCMSP、BATMAN-TCM数据库筛选三虫半夏白术天麻汤中有效活性成分及其作用靶点,并构建“中药-活性成分-交集靶点”网络图;利用GeneCard、OMMI数据库获取ICVD相关疾病靶点;将疾病靶点与成分靶点取交集,并绘制韦恩图;结合STRING数据库构建蛋白质相互作用(PPI)网络筛选核心靶点;利用David数据库进行GO和KEGG富集分析,获取潜在信号通路,并利用微生信平台进行可视化分析。结果 筛选得到三虫半夏白术天麻汤活性成分178种,靶点970个,获得ICVD相关靶点810个,三虫半夏白术天麻汤与ICVD共表达靶点165个。主要核心靶点为肿瘤坏死因子(TNF)、AKT丝氨酸/苏氨酸激酶1(AKT1)、白细胞介素-1β(IL-1β)、胰岛素(INS)、信号转导和转录激活因子3(STAT3),度值排名前5的槲皮素、山奈酚、草酸铵、β-谷甾醇、麦角胺为三虫半夏白术天麻汤潜在活性成分。GO及KEGG富集分析结果表明三虫半夏白术天麻汤主要通过流体剪切应力与动脉粥样硬化通路(Fluid shear stress and atherosclerosis)、脂质与动脉粥样硬化通路(Lipid and atherosclerosis)、糖尿病并发症AGE-RAGE信号通路(AGE-RAGE signaling pathway in diabetic complications)等信号通路发挥治疗ICVD的作用。结论三虫半夏白术天麻汤通过多成分、多靶点、多通路发挥治疗ICVD的作用。

【Abstract】 Objective To explore the mechanism of action of Sanchong Banxia Baizhu Tianma Decoction in the treatment of Ischaemic Cerebrovascular Disease(ICVD) through network pharmacology.Methods Combining TCMSP and BATMAN-TCM databases, the active ingredients and their targets in Sanchong Banxia Baizhu Tianma Decoction were screened, and constructed a network diagram of “Traditional Chinese Medicine-Active Ingredients-Targets”.GeneCard and OMMI databases were used to obtain disease targets related to ICVD.Using Venn diagram 2.1,the disease targets with the component targets were intersected and a Venn diagram was created.Protein interaction(PPI) network was constructed in combination with STRING database to screen core targets.GO and KEGG enrichment analyses were performed using the David database to obtain potential signaling pathways and visualized using the Microbiotics platform.Results A total of 178 active components and 970 targets of Sanchong Banxia Baizhu Tianma Decoction were identified, along with 810 targets related to ICVD,revealing 165 co-expressed targets between Sanchong Banxia Baizhu Tianma Decoction and ICVD.The main core targets identified were TNF,AKT1,IL-1β,INS,and STAT3.The top five ranked active components based on degree were Quercetin, Kaempferol, Ammonium oxalate, β-sitosterol, and Ergotamine, which are potential active components of Sanchong Banxia Baizhu Tianma Decoction.GO and KEGG enrichment analysis results indicate that Sanchong Banxia Baizhu Tianma Decoction exerts its therapeutic effects on ICVD primarily through signaling pathways such as Fluid shear stress and Atherosclerosis, Lipid and Atherosclerosis, and AGE-RAGE signaling pathway in diabetic complications.Conclusion The Sanchong Banxia Baizhu Tianma Decoction exerts therapeutic effects on ICVD through through multi-components, multi-targets, and multi-pathways.
