非洲马瘟病毒和西尼罗病毒多酶恒温核酸快速扩增检测方法的建立
Establishment of MIRA Detection Method for African Horse Sickness Virus and West Nile Virus
【摘要】 为了建立一种方便快捷的非洲马瘟病毒(AHSV)和西尼罗病毒(WNV)的检测方法,将GenBank中非洲马瘟病毒VP7序列以及近3年世界范围内流行的西尼罗病毒序列进行比对,选择保守区域,并按照需求设计引物与探针。通过反应条件的优化,以及特异性与敏感性试验,建立了一种能够同时检测非洲马瘟病毒和西尼罗病毒的多酶恒温核酸快速扩增(MIRA)方法。结果表明该方法操作简便、灵敏度高、特异性强,反应时间仅需20 min。该检测方法可应用于非洲马瘟和西尼罗热的快速诊断和现场监测,为疫情控制和流行病学研究提供技术支持。
【Abstract】 In order to establish a convenient and fast detection method for African horse sickness virus and West Nile virus.Compared the African horse sickness virus VP7 sequence and the West Nile virus sequence that has been circulating worldwide in the past three years in GenBank, the conservative regions were selected to design primers and probes according to requirements.A multienzyme isothermal rapid amplification(MIRA) method was established to simultaneously detect African horse sickness virus and West Nile virus through optimization of reaction conditions and specificity and sensitivity tests.The experimental results show that this method is easy to operate, highly sensitive, and has strong specificity, with a reaction time of only 20 minutes.This detection method can be applied to the rapid diagnosis and on-site monitoring of African horse sickness and West Nile fever, providing technical support for epidemic control and research.
