FGF-21凝胶联合固体微针对咪喹莫特诱导小鼠银屑病治疗的作用
Therapeutic effect of FGF-21 gel combined with solid micro targeting on imiquimod induced psoriasis in mice
【摘要】 为探究成纤维细胞生长因子21(FGF-21)凝胶联合固体微针经透皮给药治疗银屑病的可行性及作用机制,通过咪喹莫特(IMQ)诱导建立BALB-c小鼠银屑病模型,采用HE染色、荧光定量PCR、免疫组化、免疫印迹方法检测FGF-21透皮给药对银屑病的治疗效果。结果表明:FGF-21凝胶联合微针给药可显著改善银屑病样皮损,减少表皮增厚(P<0.001)和炎症细胞浸润。FGF-21可抑制p-STAT3、IL-6蛋白表达量,抑制TNF-α、IL-22、IL-17、IL-6等促炎因子mRNA转录水平,同时上调抗炎因子IL-10的mRNA转录水平。此外,FGF-21疗效呈剂量依赖性,且治疗效果优于阳性对照药他扎罗汀凝胶。携带穿膜肽R11并未增强FGF-21的治疗效果。FGF-21凝胶联合微针作为新型治疗策略,有望为银屑病患者提供安全、有效的治疗选择。
【Abstract】 To explore the feasibility and mechanism of fibroblast growth factor 21(FGF-21) gel combined with solid microneedles in the treatment of psoriasis. In this study, the psoriasis model of BALB/c mice were induced by imiquimod(IMQ). The therapeutic effect of FGF-21 on psoriasis was detected by HE staining, fluorescent quantitative PCR, immunohistochemistry and Western blotting. The results showed that FGF-21 gel combined with microneedle administration could significantly improve psoriasis like lesions, reduced epidermal thickening(P<0.001) and attenuated inflammatory cell infiltration.FGF-21 inhibited the expression of p-STAT3 and IL-6 protein, inhibited the m RNA transcription of proinflammatory factors, such as TNF-α, IL-22, IL-17, IL-6 mRNA and increased the mRNA transcription of anti-inflammatory factor IL-10. In addition, the efficacy of FGF-21 was dose-dependent, and the therapeutic effect was better than that of the positive control drug tazarotene gel. However, the incorporation of the cell-penetrating peptide R11 failed to enhance the therapeutic efficacy of FGF-21.As a new treatment strategy, FGF-21 gel combined with microneedle was expected to provide a safe and effective treatment option for patients with psoriasis.
