【摘要】 目的:分析鼻敏康合剂对变应性鼻炎(AR)最轻持续性炎症(MPI)小鼠鼻黏膜水通道蛋白(AQP)1、AQP5、黏蛋白(MUC)5AC、MUC5B的影响。方法:选取60只SPF级BALB/c雌性小鼠,适应性喂养1周后,随机分成空白组、AR组、MPI组、MPI中药组及MPI西药组,每组12只。除空白组外均以卵清白蛋白诱导、激发构建AR和MPI模型。成功后分别实施氯雷他定、鼻敏康合剂治疗方案,记录行为学积分;HE染色观察鼻黏膜形态;RT-qPCR及Western Blot、免疫组化法分别检测鼻黏膜AQP1、AQP5、MUC5AC、MUC5B mRNA及蛋白表达情况。结果:与空白组比较,MPI组鼻黏膜AQP1、MUC5AC、MUC5B蛋白及mRNA表达显著增加(P<0.01),AQP5mRNA及蛋白表达量显著降低(P<0.05,P<0.01)。与MPI组比较,两给药组鼻黏膜AQP1、MUC5AC、MUC5B蛋白及mRNA表达降低(P<0.01),AQP5 mRNA及蛋白表达上升(P<0.05,P<0.01)。结论:鼻敏康合剂可通过下调AQP1、MUC5AC、MUC5B表达,提高AQP5表达,降低鼻黏膜血管及腺上皮细胞通透性,缓解水液分泌亢进,起到治疗MPI的作用。

【Abstract】 Objective: To analyze the effects of Biminkang Mixture on nasal mucosa aquaporin(AQP)1, AQP5, mucin(MUC)5AC and MUC5B in mice with the minimal persistent inflammation(MPI) of allergic rhinitis(AR). Methods: A total of 60SPF-grade BALB/c female mice were randomly divided into blank group, AR group, MPI group, MPI Chinese medicine group and MPI Western medicine group after one week of adaptive feeding, 12 mice in each group. In addition to the blank group, the models of AR and MPI were constructed by ovalbumin induction and stimulation. After successful treatment, loratadine and Biminkang Mixture were respectively administered, and behavioral scores were recorded. The morphology of nasal mucosa was observed by HE staining. RT-qPCR, Western Blot and immunohistochemistry were used to detect the mRNA and protein expression of AQP1, AQP5, MUC5AC and MUC5B in nasal mucosa. Results: Compared with blank group, the protein and mRNA expressions of AQP1, MUC5AC and MUC5B in nasal mucosa of MPI group were significantly increased(P<0.01),the mRNA and protein expressions of AQP5 were decreased(P<0.05, P<0.01). Compared with MPI group, the protein and mRNA expressions of AQP1, MUC5AC, MUC5B in nasal mucosa of MPI Chinese medicine group and MPI Western medicine group were decreased(P<0.01), the mRNA and protein expressions of AQP5 were increased(P<0.05, P<0.01). Conclusion:Biminkang Mixture can reduce the expression of AQP1, MUC5AC and MUC5B, increase the expression of AQP5, reduce the permeability of nasal mucosal blood vessels and glandular epithelial cells, relieve hypersecretion of water, and play a role in the treatment of MPI.
