HPLC法测定人血浆、脑脊液中美罗培南的浓度
Determination of Meropenem Concentration in Human Plasma and Cerebrospinal Fluid by HPLC Method
【摘要】 目的:应用高效液相色谱法(HPLC)技术建立人血浆、脑脊液美罗培南的检测方法,为临床精准用药提供参考依据。方法:30μL样品上样于Waters sunfire C18色谱柱,以乙腈:30.0μmol·L-1乙酸铵(梯度洗脱)为流动相,分析时间15 min、检测波长300 nm,行HPLC检测。结果:血浆和脑脊液,线性范围1.0~64.0μg·m L-1、0.5~25.0μg·mL-1;回收率94.54%~111.91%、90.32%~101.86%;二者样品日内、日间精密度相对标准偏差(RSD)均<7.88%,且冻融循环检测3次均稳定。结论:本研究建立的人血浆、脑脊液美罗培南HPLC检测方法线性范围宽、准确度好、精密度高,可适用于临床治疗的常规药物监测。
【Abstract】 Objective The aim of this study was to establish an HPLC method for determining the concentration of meropenem in human plasma and cerebrospinal fluid, providing assistance for precise clinical medication. Methods 30 μL of the sample was loaded onto a Waters sunfire C18 column, using acetonitrile: 30.0 μmol·L-1 ammonium acetate(gradient elution) as the mobile phase. The analysis time was 15 minutes, and the detection wavelength was 300 nm. HPLC detection was performed. Results The plasma concentration was linear within the concentration range of 1.0 ~ 64.0 μg·mL-1, with cerebrospinal fluid concentration was linear within the concentration range of 0.5 ~ 25.0 μg·mL-1. The recovery rate of plasma quality control sample extraction was between 94.54 % ~ 111.91 %, and the recovery rate of cerebrospinal fluid quality control sample extraction was between 90.32 % ~ 101.86 %. Plasma concentration and cerebrospinal fluid concentration related standard deviation(RSD) of intra-day and inter-day assays were all less than 7.88 %, and it remained stable after three freeze-thaw cycles. Conclusions The HPLC detection method for meropenem in human plasma and cerebrospinal fluid established in this study has a wide linear range, good accuracy, and high precision, and can be applied to routine drug monitoring in clinical treatment.
