固定矫治初期唾液微生物的组成及功能变化
Microbial changes of saliva during initial stage of fixed orthodontic treatment
【摘要】 目的 探索固定矫治患者治疗初期唾液微生物的组成及功能变化。方法 纳入15名固定矫治患者纵向观察6个月,分别在初诊(BL)、洁治及口腔卫生指导(OH)、正畸3个月(TM)、正畸6个月(SM)四个时间点采集唾液,利用宏基因组测序进行物种组成、核心基因集构建及代谢通路等分析。结果 摩氏摩根菌、短隐杆菌、人心杆菌在TM组丰度最高,副干酪乳杆菌在TM组丰度最低,这四种细菌均在3个月时(TM组)丰度变化最大。经拼接组装,共获得7877076个非冗余基因,其中2044803个基因为四组共有,为“核心基因集”,占总基因783.5数的25.96%。感染性疾病和糖代谢两个KEGG二级分类在TM组明显富集。结论 在治疗前3个月物种组成及代谢功能均不稳定,3个月后有达到新平衡的趋势。
【Abstract】 Objective To explore the composition and functional changes of salivary microbiota in patients with fixed appliances during the early stage of orthodontic treatment. Methods This study included 15 patients with fixed appliances, which were longitudinally observed for 6 months. Saliva samples were collected at four time points: initial diagnosis(BL), oral hygiene instructions(OH), 3 months of orthodontic treatment(TM), and 6 months of orthodontic treatment(SM). Metagenomic sequencing was used to analyze microbial composition; core gene set construction, and metabolic pathways. Results The abundance of Morganella morganii, Cryptobacterium curtum and Cardiobacterium hominis peaked at the TM group, while the abundance of Lacticaseibacillus paracasei was the lowest at the TM group. These four bacteria showed the largest changes in abundance at 3 months(TM group), and the differences were statistically significant. After assembly, a total of 7877076 non-redundant genes were obtained, including 2044803 genes shared among all four groups,named as the “core gene set”, accounting for 25.96% of the total genes. Two KEGG secondary classifications of infectious diseases and carbohydrate metabolism were significantly enriched in the TM group. Conclusions The species composition and metabolic function are unstable during the first 3 months of treatment, but there is a trend to reach a new balance after 3months.
