朝鲜白头翁叶绿体基因组密码子偏好性分析

【摘要】 以朝鲜白头翁叶绿体基因组为研究对象,运用生物信息学方法分析其密码子使用偏好及模式,以明确其密码子使用的偏好特征,为该物种叶绿体基因工程中外源基因的密码子优化及表达水平的提高提供参考.研究发现,朝鲜白头翁叶绿体基因组密码子三个位置的碱基组成呈不均衡性,RSCU分析筛选确定16个最优密码子且其中15个以A/U碱基结尾,这一结果表明朝鲜白头翁叶绿体基因组密码子倾向于使用A/U碱基结尾.此外,ENC值及其相关性分析表明,朝鲜白头翁叶绿体基因组密码子偏好性较弱,且密码子第3位碱基GC含量变化对其偏好性的影响大于其他两个位置.

【Abstract】 The chloroplast genome of Pulsatilla cernua was investigated,and its codon usage preferences and patterns were analyzed using bioinformatics approaches. This study aimed to elucidate the characteristic features of codon usage bias in the chloroplast genome and provide insights for optimizing and enhancing the expression of exogenous genes in chloroplast genetic engineering of this species.Through RSCU analysis,16optimal codons were identified,with 15 of them ending in A/U bases. These findings suggest that the chloroplast codons of Pulsatilla cernua predominantly favor A/U endings.Furthermore,ENC values and their correlation analyses revealed a weak codon usage bias in the chloroplast genome.The GC content variation at the third codon position exerted a more significant influence on codon bias compared to variations at the other two positions.
