三相萃取葫芦巴活性部位及抗菌活性研究

【摘要】 目的:通过三相萃取葫芦巴活性部位,并对其抗菌活进行考察。方法:将石油醚、乙腈、氯仿、水以不同配比组成三相萃取体系,萃取制备葫芦巴活性部位,用牛津杯法考察抗菌活性,并用二倍稀释法测定大肠杆菌和金黄色葡萄球菌的最小抑菌浓度及最小杀菌浓度。结果:石油醚、乙腈、氯仿、水以体系1(5∶5∶1∶5)和体系2(5∶4∶2∶5)所得三相萃取物及乙醇提取物7个活性部位,与空白对照组相比,F1组对金黄色葡萄球菌有抑制作用,F2~F5组对大肠杆菌和金黄色葡萄球菌均呈抑菌作用,其中体系1下层(F3)为最佳抑菌部位,对大肠杆菌的抑制效果较好。F3对大肠杆菌的最低抑菌浓度和最小杀菌浓度分别为62.5 mg/mL和125 mg/mL;对金黄色葡萄球菌的最低抑菌浓度为125 mg/mL,无杀菌作用。结论:三相萃取可有效富集葫芦巴抗菌活性部位,效果明显。

【Abstract】 Objective: Study on active sites and antibacterial activity of fenugreek were prepared by three-phase extraction. Methods: The three-phase extraction system was composed with different ratios of petroleum ether, acetonitrile, chloroform and water, and the active sites were prepared. The antibacterial activity was investigated by Oxford cup method, and the minimum inhibitory concentration and minimum bactericidal concentration of Escherichia coli and Staphylococcus aureus were determined by two-fold dilution method. Results: Petroleum ether, acetonitrile, chloroform, water to system 1(5 ∶ 5 ∶ 1 ∶ 5) and system 2(5 ∶ 2 ∶ 5 ∶ 2) extract and ethanol extract 7 active sites.Compared with the blank control group, the F1 group inhibited S. aureus, The F2 to F5 group inbibited both E. coli and S. aureus. The lower layer of system 1 was the best antibacterial site, with good inhibition effect on E. coli. The minimum inhibitory concentration and minimum bactericidal concentration for E. coli were 62.5 mg/mL and 125 mg/mL, respectively. And the minimum inhibitory concentration for S. aureus was 125 mg/mL, with no bactericidal effect. Conclusion: Three-phase extraction could effectively enrich the antibacterial active sites of fenugreek with obvious effect.
