荨麻总黄酮提取工艺的优化及体外抗氧化活性研究

【摘要】 目的:采用超声辅助低共熔溶剂(DES)法对荨麻总黄酮的提取工艺进行研究,并探究其体外抗氧化活性。方法:以荨麻总黄酮得率为指标,确定DES的组成比例,以含水量、料液比、超声时间和超声温度为考察因素,通过单因素和Box-Behnken响应面试验,优化超声辅助DES法提取荨麻总黄酮的工艺;测定荨麻总黄酮对ABTS、DPPH以及羟基自由基的清除能力,考察其抗氧化活性。结果:当DES含水量为30%、料液比为1∶30 (g/mL)、超声时间为41 min、超声温度为61℃时,荨麻总黄酮得率为45.25 mg/g;当荨麻总黄酮提取液浓度为1.0 mg/mL时,对ABTS、DPPH、羟基自由基的清除能力分别为99.68%、52.01%和90.10%。结论:超声辅助DES法提取荨麻总黄酮的工艺稳定可靠,荨麻总黄酮具有一定的抗氧化活性。

【Abstract】 Objective: To optimize the extraction process of total flavonoids from Urtica fissa by the ultrasound-assisted deep eutectic solvent(DES) method and explore its in vitro antioxidant activity.Methods: Using the total flavonoid yield of U. fissa as the indicator, determine the composition ratios of DES. The ultrasonic-assisted DES extraction process of total flavonoids from U. fissa was optimized through single factor and Box-Behnken response surface experiments, considering factors such as water content, solid-liquid ratio, ultrasound time, and ultrasound temperature. The scavenging ability of total flavonoids of U. fissa against ABTS, DPPH, and hydroxyl radicals was determined to investigate their antioxidant activity. Results: The yield of total flavonoids from U. fissa was 45.25 mg/g at a DES water content of 30%, a solid-liquid ratio of 1∶30(g/mL), an ultrasonic time of 41 min, and an ultrasonic temperature of 61℃. When the concentration of total flavonoids extracted from U. fissa was 1.0 mg/mL, its scavenging abilities of ABTS, DPPH, and hydroxyl radicals were 99.68%, 52.01%, and 90.10%, respectively. Conclusion : The ultrasound-assisted DES method for extracting total flavonoids from U. fissa was stable and reliable, and the total flavonoids from U. fissa had certain antioxidant activity.
