滨海白首乌多糖的提取工艺研究

【摘要】 目的:优化滨海白首乌多糖的提取工艺条件,探究不同产地的白首乌多糖得率的差异,为其深度开发应用提供参考。方法:考察液料比、粉碎程度、提取温度、提取次数和提取时间对滨海白首乌多糖得率的影响,并结合响应面试验设计得出最优提取工艺条件。在最优提取工艺条件下测定不同产地白首乌多糖得率。结果:最优工艺条件为液料比22 mL/g,提取温度92℃,提取时间2.7 h,在最优工艺条件下实际测得的多糖得率为5.316%±0.018%,与模型预测值5.382%基本吻合。在该条件下提取了11个不同产地的白首乌多糖,其中江苏省盐城市滨海县所产白首乌的多糖得率为5.218%±0.049%,高于其他产地。结论:优化后的滨海白首乌多糖提取工艺稳定可靠,在所得最优工艺条件下,比较了11个不同产地的白首乌多糖得率,各产地白首乌多糖得率存在一定差异,其中江苏省盐城市滨海县所产白首乌多糖得率最高,可以作为多糖进一步开发利用的原料。

【Abstract】 Objective: To optimize the extraction conditions of polysaccharide from Binhai Baishouwu, explore the differences in the yield of polysaccharides from different habitats, and provide reference for its further development and application. Methods: The effects of liquid to material ratio, degree of pulverization, extraction temperature, extraction times and extraction time on the yield of polysaccharides from Binhai Baishouwu were investigated and the optimum extraction conditions were obtained by response surface design. The polysaccharide yield of Binhai Baishouwu from different origins were measured under the optimal extraction conditions. Results: The optimum technological conditions were 22 mL/g, 92°C, 2.7 h, and the actual yield of polysaccharide was 5.316% ± 0.018% under the optimum technological conditions, which was consistent with the predicted value of 5.382%. Under this condition, polysaccharide was extracted from 11 different habitats, and the yield of polysaccharide in Yancheng Binhai County was 5.218% ± 0.049%, which was higher than that of other regions. Conclusion: The optimized extraction process was stable and reliable. Under the optimum conditions, the yield of polysaccharide was compared in 11 different habitats. There were some differences in the yield of polysaccharide from different habitats, the highest yield of polysaccharide was found in Yancheng Binhai County and it can be used as raw material for further development and utilization of polysaccharide.
