miR-3189-3p对结直肠癌患者预后的影响及作用机制探讨
An exploration on the effect of miR-3189-3p on prognosis of patients with colorectal cancer and its mechanism of action
【摘要】 目的 探讨miR-3189-3p对结直肠癌(CRC)患者预后的影响及作用机制。方法 基于CancerMIRNome数据库和ENCORI数据库数据分析miR-3189-3p在CRC组织和正常组织中的表达差异,探讨miR-3189-3p表达水平对患者生存预后的影响。分别将hsa-miR-3189-3p mimics、miR-3189-3p NC转染至LOVO细胞,通过Western blot实验探讨miR-3189-3p对LOVO细胞上皮细胞-间充质转化(EMT)关键蛋白[波形蛋白(vimentin)和N钙黏蛋白(N-cadherin)]表达水平的影响,通过划痕实验和Transwell实验探讨miR-3189-3p对LOVO细胞迁移和侵袭能力的影响。结果 CRC组织的miR-3189-3p表达水平显著高于正常组织(P<0.05)。miR-3189-3p高表达CRC患者的生存预后较低表达者更好(P<0.05)。过表达miR-3189-3p后,LOVO细胞的迁移和侵袭能力显著减弱(P<0.05),vimentin和N-cadherin蛋白表达水平显著降低(P<0.05)。结论 miR-3189-3p可能通过调控CRC细胞的迁移、侵袭以及EMT对患者的生存预后造成影响。
【Abstract】 Objective To explore the effect of micro ribonucleic acid-3189-3p(miR-3189-3p) on prognosis of patients with colorectal cancer(CRC) and its mechanism of action.Methods Based on the data of CancerMIRNome database and ENCORI database, the differences in the expressions of miR-3189-3p in CRC tissues and normal tissues were analyzed, and the effect of the level of miR-3189-3p expression on the survival prognosis of the patients was explored. Hsa-miR-3189-3p mimics and miR-3189-3p NC were transfected into LOVO cells, respectively. The effects of miR-3189-3p on the expression levels of the key proteins(vimentin and N-cadherin) of epithelial-mesenchymal transition(EMT) in the LOVO cells were explored by using Western blot assay. The effects of miR-3189-3p on the migration and invasion ability of the LOVO cells were explored by using scratch assay and Transwell assay.Results The level of miR-3189-3p expression in the CRC tissues was significantly higher than that in the normal tissues(P<0.05). The survival prognosis of the CRC patients with high expression of miR-3189-3p was better than that of the CRC patients with low expression of miR-3189-3p(P<0.05). After overexpression of miR-3189-3p, the migration and invasion ability of the LOVO cells were significantly weakened(P<0.05), and the expression levels of vimentin and N-cadherin proteins were significantly decreased(P<0.05).Conclusion MiR-3189-3p may affect the survival prognosis of the patients by regulating the migration, invasion and EMT of CRC cells.
