黄芪中有效成分含量检测方法的建立
Establishment of a method for determining the content of active components in Radix Astragali
【摘要】 《中华人民共和国药典》中采用不同的黄芪样品制备方法和不同的色谱方法分别检测毛蕊异黄酮葡萄糖苷和黄芪甲苷。本研究拟对样品制备方法和两种成分的检测方法进行合并统一,利用高效液相色谱建立黄芪中有效成分的含量测定方法。采用Waters Atlantis?T3色谱柱(250.0 mm×4.6 mm,5.0μm),以水和乙腈为流动相,梯度洗脱,流速1 mL/min,色谱柱温度30℃,检测波长260 nm,增益值1 000,漂移管温度65℃,空气压力30 psi,喷雾器温度36℃。结果显示,毛蕊异黄酮葡萄糖苷和黄芪甲苷标准品在各自范围内线性关系良好(r>0.999 0);毛蕊异黄酮葡萄糖苷的平均回收率和相对标准偏差分别为97.66%(n=6)和1.45%,黄芪甲苷的平均回收率和相对标准偏差分别为99.64%(n=6)和0.38%。18批黄芪中2种成分的含量分别为0.366 3~1.610 1 mg/mL和0.473 1~1.666 0 mg/mL。本方法缩短了检测时间,提高了检测效率,且快速可靠,稳定性和重复性好,可用于黄芪的质量控制。
【Abstract】 Radix Astragalisamples were prepared by different preparation methods in Chinese Pharmacopoeia, and then different chromatographic methods were used to detect the calycosin glycoside and astragaloside Ⅳ, respectively.Based on this design experiment, the preparation method and detection method of the two components were combined and unified, to establish the same preparation method and the same chromatographic method for the determination of active components in Radix Astragali by high performance liquid chromatography. The chromatography was performed on Waters Atlantis?T3 column(250.0 mm×4.6 mm, 5.0 μm) with water and acetonitrile as mobile phase and gradient elution. The flow rate was 1 mL/min. The column temperature was 30℃. The detection wavelength was 260 nm. Gain value: 1 000; drift tube temperature: 65℃, air pressure: 30 psi, sprayer temperature: 36℃. The results showed that the linear relationship between the standard products of calycosin glucoside and astragaloside Ⅳ was good in their respective ranges(r>0.999 0). The average recovery and relative standard deviation of calycosin glucoside were 97.66%(n=6) and1.45%, and the average recovery and relative standard deviation of astragaloside were 99.64%(n=6) and 0.38%. The contents of 2 components in 18 batches of Radix Astragali were 0.366 3-1.610 1 mg/mL and 0.473 1-1.666 0 mg/mL,respectively. The established method is fast, reliable, stable and reproducible, and can be used for the quality control of Radix Astragali.
