基于糖谱的抗衰老口服液质量评价研究
Research on the quality evaluation of anti-aging oral liquid based on saccharide mapping
【摘要】 利用液相色谱分析技术,建立糖谱,对抗衰老口服液中糖类物质进行质量控制,并结合化学模式识别,对配制过程工艺参数进行分析。采用HPLC,色谱柱为Shodex Asahipak NH2P-50 4E(4.6 mm×250 mm, 5.0μm),流动相为纯水-乙腈,梯度洗脱,ELSD漂移管温度为100℃;氮气流速为1.6 mL/min的液相条件,对抗衰老口服液及配制液的糖类物质进行定性和定量分析。结果表明:抗衰老口服液及配制液中含有D-(-)-阿拉伯糖、D-果糖、D-(+)-甘露糖、D-葡萄糖、蔗糖、蜜二糖、D-棉籽糖、甘露三糖和水苏糖等,寡糖及单糖的主要来源为生地黄,其中D-果糖、D-葡萄糖、甘露三糖、水苏糖定量方法学考察良好。化学模式识别分析结果显示:5℃放置3 d寡糖及单糖变化显著,5~7 d较为稳定;溶液pH为3.5~4.5时变化趋势较稳定。基于OPLS-DA模型筛选出VIP值大于1的成分为D-棉籽糖、D-(-)-阿拉伯糖、甘露三糖以及D-果糖,这些成分可作为配制工艺过程的关键化学标志物。
【Abstract】 A saccharide mapping method was established for quality control of carbohydrate substances in anti-aging oral liquid using liquid chromatography analysis technology. The process parameters of the formulation were analyzed by combining the saccharide mapping with chemometric pattern recognition. The chromatographic analysis was performed using HPLC with a Shodex Asahipak NH2P-50 4E(4.6 mm×250 mm, 5.0 μm). The mobile phase consisted of a gradient elution of pure water and acetonitrile. The ELSD drift tube temperature was set to 100 ℃; and the nitrogen flow rate was 1.6 mL/min. Qualitative and quantitative analysis of anti-aging oral liquid and its preparation solutions were conducted under these chromatography conditions. The results showed that serval sugars in the anti-aging oral liquid, including D-(-)-arabinose, D-fructose, D-(+)-mannose, D-glucose, sucrose, melibiose, D-raffinose, manninotriose, and stachyose. The primary source of the oligosaccharides and monosaccharide was raw Rehmannia glutinosa. The quantitative methodology of D-fructose, D-glucose, manninotriose, and stachyose was well evaluated. Chemical pattern recognition analysis revealed significant changes in oligosaccharides and monosaccharide after 3 days of storage at 5 ℃. However, the solution remained relatively stable after 5 to 7 days. The pH of the solution stabilized between 3.5 to 4.5. Based on the OPLS-DA model, components with VIP values greater than 1 were selected as D-raffinose, D-(-)-arabinose, manninotriose, and D-fructose, which can serve as key chemical markers in the preparation process of anti-aging oral liquid.
