巴戟天-丹参对去卵巢大鼠骨质疏松影响的研究

【摘要】 目的 探讨巴戟天-丹参配伍对去卵巢大鼠骨质疏松的影响。方法 选取SD雌性大鼠30只,随机分为假手术组、模型组、戊酸雌二醇治疗组、巴戟天-丹参(低、中、高剂量)治疗组。除假手术组外,其余各组均采用去卵巢手术诱导骨质疏松模型,灌胃给药12周后取材。测定各组血清骨转化指标,进行股骨Micro-CT检测和骨微结构分析,观察并分析股骨的HE及TRAP染色结果。结果 血清骨转化结果显示,与模型组相比,巴戟天-丹参治疗组及戊酸雌二醇治疗组的血清OPG、PINP、OCN、BMP-2水平均升高(P<0.01),TRACP、β-CTX水平均降低(P<0.01);Micro-CT结果显示,与模型组相比,巴戟天-丹参治疗组及戊酸雌二醇治疗组的骨密度(BMD)、骨体积/总体积(BV/TV)升高(P<0.01),骨表面积/骨体积(BS/BV)降低(P<0.01),皮质骨厚度(Ct.Th, mm)、皮质骨面积/骨总面积(Ct.Ar/Tt.Ar,%)升高(P<0.01),骨小梁厚度(Tb.Th, mm)及骨小梁数量(Tb.N,mm-1)增加(P<0.01),骨小梁分离度(Tb.Sp, mm)降低(P<0.01);HE染色及RAP染色结果显示,与模型组相比,巴戟天-丹参治疗组及戊酸雌二醇治疗组的骨小梁结构、数目、连续性均有明显改善,破骨细胞数量减少,且巴戟天-丹参中剂量治疗组效果优于戊酸雌二醇治疗组。结论 巴戟天-丹参配伍使用,可明显改善骨质疏松大鼠血清骨转化指标,改善骨微观结构并增加骨密度,对去卵巢大鼠的骨质疏松有显著的保护性作用,且中剂量组治疗效果优于戊酸雌二醇治疗组。

【Abstract】 Objective To explore the effect of morinda officinalis-salvia miltiorrhiza combination on osteoporosis in ovariectomised rats. Methods Thirty SD female rats were randomly divided into sham operation group, model group, estradiol valerate treatment group, and morinda officinalis-salvia miltiorrhiza(low, medium, and high dose) treatment group. Except for the sham-operated group, ovariectomy was performed in the rest of the groups to induce osteoporosis model. Rats were collected after 12 weeks of gavage administration. Serum bone transformation indexes were measured in each group, as well as femur micro-CT detection and bone microstructure analysis. HE and TRAP staining result of the femur were also observed and analyzed. ResultsCompared to those in the model group, the result of serum bone transformation showed that serum levels of OPG, PINP, OCN, and BMP-2 elevated(P<0.01), but levels of TRACP and β-CTX decreased(P<0.01) in the morinda officinalis-salvia miltiorrhiza treatment group and the estradiol treatment group. Meanwhile the result of micro-CT showed that bone mineral density(BMD), bone volume/total volume(BV/TV) increased(P<0.01), bone surface area/bone volume(BS/BV) decreased(P<0.01), cortical bone thickness(Ct.Th, mm) and cortical bone area/total bone area(Ct.Ar/Tt. Ar, %) increased(P<0.01), trabecular thickness(Tb.Th, mm) and number of trabeculae(Tb.N, mm-1 )increased(P<0.01), and trabecular separation(Tb.Sp, mm) decreased(P<0.01) in the morinda officinalis-salvia miltiorrhiza treatment group and the estradiol treatment group. The result of HE staining and RAP staining showed that compared to those in the model group, the structure, number, and continuity of bone trabeculae were significantly improved and the number of osteoclasts reduced in the morinda officinalis-salvia miltiorrhiza treatment group and the estradiol treatment group. Simultaneously, the effect in morinda officinalis-salvia miltiorrhiza medium-dose treatment group was better than that in the estradiol treatment group. Conclusion The addition of morinda officinalis-salvia miltiorrhiza in combination significantly improves the bone conversion index and bone microstructure, and increases bone density in osteoporotic rats, which has a significant protective effect against osteoporosis in ovariectomized rats much better than estrogen treatment.
