山茶刺盘孢LAMP可视化快速检测方法的建立

【摘要】 为实现对茶炭疽病的快速诊断,针对其主要病原菌山茶刺盘孢(Colletotrichum camelliae)建立LAMP检测技术体系。以山茶刺盘孢ApMat基因片段设计特异性引物,验证其特异性,优化反应时间和温度,测试反应的灵敏度,并在田间病叶上进行了应用验证。结果表明,在25μL的LAMP检测反应体系中,最适反应温度为63℃,最佳反应时间60 min;最佳反应条件下检测灵敏度可达100 pg/μL,由此构建的山茶刺盘孢LAMP检测体系能够特异性检测山茶刺盘孢,可应用于田间病叶的早期诊断。

【Abstract】 A rapid detection system based on the loop-mediated isotherm al amplification(LAMP) was established for Colletotrichum camelliae, the main pathogen of tea anthracnose. The ApMat gene fragment of C. camelliae was used to design specific primers, and the specificity of the primers was verified. The reaction time and temperature were optimized,and the sensitivity of the reaction was tested. In the 25 μL LAMP detection reaction system, the optimum reaction temperature was identified as 63 ℃ and the optimum reaction time as 60 minutes. Under these optimal reaction conditions, the sensitivity of the system was determined to be 100 pg/μL. The LAMP visualized rapid detection system can be used for the specific detection of C. camelliae and the early diagnosis of tea anthracnose in the field.
