哈萨克药繁枝补血草总黄酮提取分离纯化工艺及抗氧化活性研究
Study on the Extraction, Separation, Purification Process and Antioxidant Activity of Total Flavonoids from Kazakh Medicinal Limonium myrianthum(Schrenk) Kuntze
【摘要】 目的 建立繁枝补血草总黄酮的最佳提取工艺,利用大孔树脂确定总黄酮最佳纯化工艺,测定其抗氧化活性。方法 以繁枝补血草总黄酮含量为指标,选取最适宜繁枝补血草总黄酮的提取方法。根据单因素实验,正交实验选取最优提取工艺。筛选最适宜大孔树脂,确定最佳纯化工艺。利用体外抗氧化能力实验,研究总黄酮的抗氧化能力。结果 料液比1∶50 g/ml,50%乙醇加热回流60 min,提取3次,最高提取率13.77%。选用X-5型大孔树脂,样品浓度1.270 mg/ml,70%乙醇洗脱,径高比1∶10,流速2 BV/h,总黄酮纯度80.3%。DPPH清除实验显示,繁枝补血草总黄酮提取物抗氧化能力强,DPPH自由基清除率IC50值为0.8492 mg/ml。结论 提取纯化工艺可行,且提取物具有很好的抗氧化活性。
【Abstract】 Objective Establish the optimal extraction process for total flavonoids in Limonium myrianthum(Schrenk) Kuntze, determine the optimal purification process for total flavonoids using macroporous resin, and evaluate their antioxidant activity. Methods Conduct experiments using ultrasonic extraction and heated reflux extraction methods, and select the optimal extraction method based on the total flavonoid content as the indicator. Determine the optimal extraction process for total flavonoids in Limonium myrianthum(Schrenk) Kuntze through single factor experiments and orthogonal experiments. Using static adsorption rate and desorption rate as indicators, select the optimal resin from 7 types of macroporous resins and determine the optimal purification process. Study the antioxidant capacity of total flavonoids through the in vitro antioxidant capacity index method. Results The extraction ratio was 13.77% at a material-to-liquid ratio of 1∶50 g/ml using 50% ethanol with reflux heating for 60minutes, extracted three times. The X-5 macroporous resin was selected, with a sample concentration of 1.270 mg/ml, eluted with 70% ethanol, a ratio of diameter to height of 1∶10, a flow rate of 2 BV/h, and a total flavonoid purity of 80.3%. The DPPH scavenging experiment indicated that the total flavonoids extracted from Gynura divaricata had strong antioxidant capacity, with an IC50 value for DPPH radical scavenging rate of 0.849 2mg/ml. Conclusions This extraction and purification process is feasible, and the extract has good antioxidant activity.
