2024年北京市19家疾控机构病原微生物实验室建设和生物安全管理现况调查

【摘要】 目的 调查2024年北京市、区两级疾控机构病原微生物实验室建设和生物安全管理现状,为疾控机构生物安全管理和发展提出建议。方法 2024年,对北京市、区级19家疾控机构的生物安全二级(BSL-2)实验室、核酸检测实验室、高通量测序(NGS)实验室、菌毒种及生物样本库、危化品库的建设和管理以及病原微生物实验活动开展情况进行问卷调查和分析。结果 2024年北京市疾控机构共建设BSL-2实验室257间(套),其中包括负压加强型生物安全二级(BSL-2+)实验室52间(套)。除北京经济技术开发区疾病预防控制中心外,区级疾控建有BSL-2实验室3~23间(套),面积250.00~2 082.00 m~2,并开展包括培养鉴定等各类病原微生物检测活动,涉及第二类病原微生物3~10种、第三类病原微生物19~92种。各区建有核酸检测实验室1~6套,面积57.30~900.00 m~2,94.74%的real-time PCR实验室进行了合理分区,47.37%(9/19)配备了新型一体化PCR实验室,52.63%(10/19)建有NGS实验室,89.47%(17/19)拥有独立菌毒种/生物样本库或保存区域,57.89%(11/19)建有危化品库/存储间,90.91%(10/11)设专人管理危化品。生物安全检查发现实验室存在设施设备不全和使用不规范等问题。结论 2024年北京市疾控机构病原微生物实验室建设已经较完备,生物安全管理水平较高。应针对北京市疾控系统检测病原种类多、开展实验活动种类丰富、检测技术更新快的特点,加强风险评估和风险控制,同时建立针对新发突发传染病和未知病原检测等新的生物安全管理体系及标准,确保在重大传染病疫情防控、大型活动保障中高水平开展防病检测工作。

【Abstract】 Objective To investigate the construction of pathogenic microbiology laboratories and biosafety management in CDC of city level and district level in Beijing city, so as to make recommendations for biosafety management and development in CDC. Methods A questionnaire survey was conducted on the construction and management of BSL-2(Biosafety Level 2 Laboratory) laboratories, nucleic acid testing laboratories, NGS(Next Generation Sequencing) laboratories, bacterial and toxin species and biological sample banks, hazardous chemicals banks, and the experimental activities of pathogenic microorganisms among 19 CDCs in Beijing city. Results There were 257 BSL-2 laboratories(sets) in CDC of city level and district level in Beijing city, including 52 negative pressure enhanced BSL-2+ laboratories(sets).All BSL-2 laboratories have been filed.Except for Beijing Economic-Technological Development Area Disease Prevention and Control Center, there were BSL-2(3-23) rooms(sets) built at the district level CDC,covering an area of 250.00-2 082.00 m~2,various pathogenic microorganism detection activities including cultivation and identification were carried out, including 3-10 species of class Ⅱ pathogenic microorganisms and 19-92 species of Class Ⅲ pathogenic microorganisms.There were 1 to 6 nucleic acid testing laboratories at the district level CDC,covering an area of 57.30 to 900.00 m~2.About 94.74% of the real-time PCR laboratories have been properly partitioned, and 47.37%(9/19) had integrated PCR laboratories, 52.63%(10/19) have established NGS laboratories, 89.47%(17/19) had independent libraries of bacterial/biological samples and dedicated storage areas, 57.89%(11/19) of CDC institutions had specialized storage rooms for hazardous chemicals, and 90.91%(10/11) had full-time personnel to manage hazardous chemicals.The biosafety inspection found that the laboratory had inadequate facilities and equipment and improper use. Conclusion In 2024,the construction of pathogenic microorganism laboratories in Beijing′s disease control institutions has been relatively complete, and the level of biosafety management is relatively high.Based on the characteristics of a wide range of pathogen detection and diverse experimental activities conducted by Beijing′s disease control system, as well as the rapid update of detection technologies, it is necessary to strengthen risk assessment and risk control.At the same time, a new system of biological safety management and standards should be established to cover the detection of emerging and unexpected infectious diseases and unknown pathogens, in order to ensure that high-level disease prevention and detection work can be carried out during major infectious disease outbreaks and large-scale event support.
