玉米HKT基因家族全基因组鉴定及生物信息学分析
Whole Genome Identification and Bioinformatics Analysis of Maize HKT Gene Family
【摘要】 基于14个物种的HKT基因家族构建系统进化树,深入分析模式作物拟南芥及玉米、高粱、水稻3个亲缘关系较近的禾本科物种HKT家族成员的基因结构、保守基序及其蛋白质理化性质等,对玉米HKT基因家族进行全基因组鉴定和系统的生物信息学分析。分别用200 mmol/L NaCl和100 mmol/L Na2CO3对郑58(抗)和昌7-2(感)的玉米幼苗进行盐、碱胁迫处理,在胁迫0、1和3 d时取叶部及根部样品进行转录水平分析。结果表明,14个物种的HKT家族成员在系统进化树划分为两个支系,推测可能至少来自两个共同的祖先基因。HKT家族具有相似的基因结构和特殊的基因特征,玉米HKT基因家族中共有包括1个I型和两个II型在内的3个成员,推测拥有更多的HKT基因家族成员有助于植物在盐碱环境下获得更广泛的适应性。此外,盐碱处理下基因表达分析显示,郑58和昌7-2的耐盐碱性差异与ZmHKT1;1直接相关,与ZmHKT2;1间接相关,与ZmHKT1;2无关。
【Abstract】 A phylogenetic tree was constructed based on the HKT gene family of 14 species, and the gene structure, conserved motifs, and physicochemical properties of the HKT family members of the model crop Arabidopsis Thaliana and three closely related species of maize, sorghum, and rice were deeply analyzed. The alkali-tolerant inbred line Zheng58 and the alkali-sensitive inbred line Chang7-2 were used as experimental materials to transcription analysis data of the leaf and root for the seedlings under the stress of 200 mmol/L NaCl or 100 mmol/L Na2CO3solution at stress 0, 1 and 3 days. The results show that the HKT family members of 14 species are divided into two clades in the phylogenetic tree, which may be derived from at least two common ancestral genes. The HKT family has similar gene structure and special gene characteristics. There are 3 members of the maize HKT gene family,which included one type I and two type II. This paper hypothesizes that having more members of the HKT gene family contributes to a broader adaptation of plants to salt-alkali environments. Moreover, gene expression analysis under salt-alkali treatment showed that the salt-alkali tolerance of Zheng58 and Chang7-2 was directly related to ZmHKT1;1, indirectly related to ZmHKT2;1 and irrelevant to ZmHKT1;2. This study reveals the regulatory mechanism of HKT gene and has important implications for the comparative genomics analysis of HTK gene families, understanding plant development and response to abiotic stress.
