【摘要】 目的 探讨九肽-1/依克多因经皮共递送纳米脂质体(Non/Ect-NLPs)的美白修复功效。方法 通过细胞活性测试研究Non/Ect-NLPs安全性,观察黑素细胞对纳米脂质体摄取情况,并比较不同水平Non/Ect-NLPs组及游离成分溶液组酪氨酸酶活性、黑色素水平、氧化损伤细胞存活率及人源水通道蛋白-3(AQP3)、丝聚合蛋白(FLG)、紧密连接蛋白1(Claudin-1)水平。结果 游离成分溶液组和不同水平Non/Ect-NLPs组中HaCaT细胞活性均在80%以上。与游离成分溶液组比较,Non/Ect-NLPs水平为250.0、500.0μg/mL时,其对B16F10细胞酪氨酸酶活性显著降低(P<0.05)。与游离成分溶液组比较,Non/Ect-NLPs水平为125.0、500.0μg/mL时,其能显著抑制B16F10细胞分泌黑色素(P<0.05)。与模型组比较,Non/Ect-NLPs水平为250.0、500.0μg/mL时,可显著提高氧化损伤细胞的存活率(P<0.05)。与对照组比较,游离成分溶液组和Non/Ect-NLPs组能显著促进HaCaT细胞迁移(P<0.01)。与对照组比较,游离成分溶液组、Non/Ect-NLPs组AQP3、FLG、Claudin-1水平显著增高(P<0.05)。结论 Non/Ect-NLPs作为新型皮肤美白修复功效原料具有良好的应用前景。

【Abstract】 Objective To explore the whitening and repairing effects of nonapeptide-1/ectoin nanoliposomes(Non/Ect-NLPs).Methods The safety of Non/Ect-NLPs was studied by cell viability test, and the uptake of nanoliposomes by melanocytes was observed.Tyrosinase activity, melanin levels, survival rate of oxidative damage cells, and the levels of human aquaporin-3(AQP3),filaggrin(FLG),and tight junction protein-1(Claudin-1) levels were compared between groups with different levels of Non/Ect-NLPs and free component solution groups.Results The viability of HaCaT cells in both the free component solution group and the groups with different levels of Non/Ect-NLPs were all above 80%.Compared with the free component solution group, when the levels of Non/Ect-NLPs were 250.0 and 500.0 μg/mL,the tyrosinase activity of B16F10 cells was significantly decreased(P<0.05).Compared with the free component solution group, when the levels of Non/Ect-NLPs were 125.0 and 500.0 μg/mL,it could significantly inhibit the secretion of melanin by B16F10 cells(P<0.05).Compared with the model group, Non/Ect-NLPs at 250.0 and 500.0 μg/mL significantly increased the survival rate of oxidative damaged cells(P<0.05).Compared with the control group, the free component solution group and the Non/Ect-NLPs group significantly promoted the migration of HaCaT cells(P<0.01).Compared with the control group, the free component solution group and the Non/Ect-NLPs group had significant increases in AQP3,FLG,and Claudin-1 levels(P<0.05).Conclusion Non/Ect-NLPs have a good application prospect as a new type of skin whitening and repair efficacy raw material.
