2021—2022年南京市野猪中8种病毒感染情况及PCV2全基因组遗传进化分析

【摘要】 野猪是多种重要动物疫病的天然宿主,为了解2021—2022年期间南京市野猪疫病感染情况、基因型及遗传变异特征,为野猪疫病的防控提供科学依据,在江苏省南京市共采集30头野猪的76份样品,其中30份全血样品、30份血清样品、6份粪便样品和10份组织样品(心脏、肝脏、脾脏、肺脏和肾脏组织的混合样品)用于开展猪瘟病毒(CSFV)、非洲猪瘟病毒(ASFV)、猪繁殖与呼吸综合征病毒(PRRSV)、伪狂犬病病毒(PRV)、口蹄疫病毒(FMDV)、猪流行性腹泻病毒(PEDV)、猪圆环病毒2型(PCV2)和猪圆环病毒3型(PCV3)的病毒核酸及抗体检测。不同样品PCR检测结果显示,30头野猪中有12头PCV3阳性,阳性率为40%;PCV2阳性率为6.7%;其余病原核酸检测均为阴性。在PCV2检测阳性的野猪样品中成功获得2条PCV2全基因组序列,且均属于PCV2d基因亚型。血清抗体检测结果显示,野猪PCV3、PCV2、PRV gB、PRRSV、PEDV和FMDV A型抗体阳性率分别为53%、30%、30%、3.3%、3.3%和6.7%;所有野猪CSFV、ASFV、PRV gE抗体和FMDV O型抗体检测均为阴性。本研究揭示了南京市野猪携带或感染过多种重要病毒,需持续对野猪进行疫病监测,并研究野猪与家猪间的疾病传播规律。

【Abstract】 Wild boars are natural hosts for many important animal diseases. To conduct a comprehensive investigation into the prevalence, genotype, and genetic variability of wild boar diseases in Nanjing and to provide a scientific basis for effective prevention and control of these diseases in wild boar populations, 76 samples, including 30 blood, 30 serum, 6 feces and 10 tissue samples(a mixture of heart, liver,spleen, lung and kidney), from 30 wild boars in Nanjing, Jiangsu Province were collected from March 2021 to June 2022 for molecular and serological surveillance of classical swine fever virus(CSFV), African swine fever virus(ASFV), porcine respiratory and reproductive disorder syndrome virus(PRRSV), pseudorabies virus(PRV), foot-and-mouth disease virus(FMDV), porcine epidemic diarrhea virus(PEDV), porcine circovirus type 2(PCV2) and porcine circovirus type 3(PCV3). The PCR results showed that PCV3 nucleic acid was detected in 12 wild boars with a positive rate of 34%, PCV2 nucleic acid was detected in 3 wild boars with a positive rate of 6. 7%, and all the wild boars were negative for the remaining viruses. Two whole genome sequences were obtained from the PCV2-positive samples, and both sequences belong to the PCV2d genotype. The serological tests showed that the positive rates of PCV3 antibody, PCV2 antibody, PRV gB antibody, FMDV type A antibody, PRRSV antibody and PEDV antibody were 53%, 30%, 30%, 6. 7%, 3. 3% and 3. 3%, respectively.All the wild boars showed negative for PRV gE, FMDV type O, CSFV and ASFV antibodies. This study revealed that the wild boar population in Nanjing carried or had been exposed to various important swine viral pathogens. In the future, continuous surveillance of wild boar diseases and research on the transmission patterns between wild boars and domestic pigs are required.
