羊痘病毒81基因原核表达载体构建及蛋白表达

【摘要】 为研究羊痘病毒81基因的生物学功能,构建羊痘病毒81基因的原核表达载体并表达其蛋白。利用在线软件对羊痘病毒81基因进行生物信息学分析,通过PCR技术扩增羊痘病毒81基因,与原核表达载体PET-42b构建重组质粒,大肠感受态细胞表达其蛋白。结果表明:羊痘病毒81基因蛋白无信号肽,无跨膜区,有19个抗原决定簇,二、三级结构预测表明其以无规则卷曲为主,表现出疏水性;PCR成功扩增羊痘病毒81基因(492 bp),PET-42b-81原核表达载体构建成功,成功表达18.35 kDa大小81基因蛋白。本研究为深入探究羊痘病毒中间基因提供基础。

【Abstract】 To investigate the biological function of the sheep poxvirus 81 gene, a prokaryotic expression vector was constructed to expess its protein. Bioinformatics analysis of the 81 gene was conducted using online software, afollowed by gene amplification through PCR and the creation of a recombinant plasmid with the prokaryotic expression vector PET-42b. The protein was expressed in colon receptor cells. The study revealed that the 81 gene protein lacked a signal peptide and transmembrane region, contained 19 antigenic determinants, and was predicted to have an irregularly coiled, hydrophobic secondary and tertiary structure. PCR successfully amplified the 81 gene, and the PET-42b-81 prokaryotic vector was able to express the 18.35 kDa 81 gene protein. These results lay the groundwork for further exploration of the core genes of sheep poxviruses.
