天山假狼毒化学成分及抗菌活性研究
Study on Chemical Constituents and Antibacterial Activity of Stelleropsis tianschanica (Pobed)
【摘要】 目的 研究瑞香科植物天山假狼毒Stelleropsis tianschanica (Pobed)的化学成分及抗菌活性。方法 采用柱色谱的分离方法对天山假狼毒的95%乙醇提取物进行分离纯化,利用核磁共振NMR、高分辨质谱等方法,结合理化鉴定化合物结构。采用滤纸片法对不同浓度的单体化合物的抗菌活性进行筛选。结果 从天山假狼毒中分离鉴定11个化合物,分别为:(E)-11-methoxy-11-oxoundec-2-enoic acid(1)、Methyl-(E)-9-oxooctadec-10-enoate(2)、3-Hydroxyphthalimide(3)、Dimethyl hexadecanedioate(4)、Octadecane-1,18-dimethylester(5)、壬二酸氢甲酯(6)、正十六烷酸(7)、bothrioclinin(8)、Daphnolon(9)、β-谷甾醇(10)、芝麻素(11)。化合物7(128μg/ml)对金黄色葡萄球菌有一定抑制活性,抑菌圈大小为8.65 mm。化合物9(128μg/ml和64μg/ml)对鲍曼不动杆菌有抑制活性,抑菌圈大小分别为12.65 mm和8.05 mm。结论 化合物1~9为首次从天山假狼毒中分离得到;其中化合物1~3的碳谱数据为首次报道。化合物7和化合物9均表现出一定的抗菌活性。
【Abstract】 Objective To study the chemical constituents of Stelleropsis tianschanica(Pobed), a Thymelaeaceae plant.Methods Using column chromatography to separate and purify the pure methanol extract of Stelleropsis tianschanica(Pobed) and the structure of the compound was rationalized and identified using nuclear magnetic resonance NMR and high-resolution mass spectrometry.The filter paper method was used to screen the antibacterial activity of monomeric compounds at different concentrations. Results Eight compounds were obtained and identified as(E)-11-methoxy-11-oxoundec-2-enoic acid(1)、Methyl-(E)-9-oxooctadec-10-enoate(2)、3-Hydroxyphthalimide(3)、Dimethyl hexadecanedioate(4)、Octadecane-1,18-dimethylester(5)、9-methoxy-9-oxononanoic acid(6)、n-Hexadecane acid(7)、Bothrioclinin(8)、Daphnolon(9)、β-sitosterol(10)、Sesamin(11). Compound 7(128 μg/ml) had some inhibitory activity against Staphylococcus aureus, with a size of 8.65mm of the antibacterial ring, and compound 9(128 μg/ml and 64 μg/ml) had inhibitory activity against Acinetobacter baumannii, with antibacterial ring of 12.65 mm and 8.05 mm.Conclusion Compounds 1~9 were obtained from this plant for the first time and the 13C-NMR data of compounds 1~3 were reported for the first time. Compounds 7 and 9 showed certain antimicrobial activity.
