可溶性鸟苷酸环化酶刺激剂对急性心肌梗死小鼠心功能的影响
The effect of soluble guanylate cyclase stimulators on cardiac function in mice with acute myocardial infarction
【摘要】 目的 探讨可溶性鸟苷酸环化酶(sGC)刺激剂维立西呱对急性心肌梗死小鼠心功能的影响。方法 健康SPF级雄性C57BL/6J小鼠采用随机数字法分为3组:假手术组、模型组(MI)和治疗组(MI+Veri),每组8只。MI组和MI+Veri组通过左冠状动脉前降支(LAD)结扎术建立急性心肌梗死(AMI)模型。MI+Veri组在术后24h予以维立西呱5mg/kg/d灌胃;对照组及MI组给予等量生理盐水灌胃。LAD结扎术后2、4周,超声心动图随机检测各组小鼠舒张末期室间隔厚度(IVSd)、收缩末期室间隔厚度(IVSs)、左心室舒张末期容积(LVEDV),左心室收缩末期容积(LVESV),左心室舒张末期内径(LVIDd)、左心室收缩末期内径(LVIDs),同时计算左室缩短分数(LVFS)和左室射血分数(LVEF)。LAD结扎术后4周,采血进行血清生化SCr及BUN测定。结果 术后2周,与假手术组相比,MI组小鼠LVESV、LVESD升高(P均<0.05),LVEF、LEFS降低(P均<0.001);MI+Veri组小鼠无统计学差异(P均>0.05)。术后4周,与假手术组相比,MI组及MI+Veri组小鼠LVEF、LVFS均降低(P均<0.001);MI+Veri组较MI组小鼠LVESV、LVESD降低(P均<0.05)、LVEF、LVFS升高(P均<0.001)。血清生化BUN结果显示MI组较假手术组升高(P<0.05);MI+Veri组较MI组小鼠降低(P<0.05)。结论 维立西呱可在一定程度上改善急性心肌梗死小鼠急性心功能恶化,逆转心室重构。
【Abstract】 Objective To investigate the effects of soluble guanylate cyclase(sGC) stimulator Vericiguat on cardiac function in mice with acute myocardial infarction(AMI).Methods Healthy male SPF-grade C57BL/6J mice were randomly divided into three groups: sham surgery group, MI group, and MI+Veri group, with 8 mice per group. The MI and MI+Veri groups underwent left anterior descending(LAD) coronary artery ligation to establish an acute myocardial infarction(AMI) model. The MI+Veri group was administered Vericiguat at 5 mg/kg/d via gavage 24 hours post-surgery, while the control and MI groups received an equal volume of saline. At 2 and 4 weeks post-LAD ligation, echocardiography was performed to assess the following parameters: interventricular septal thickness at end-diastole(IVSd), interventricular septal thickness at end-systole(IVSs), left ventricular end-diastolic volume(LVEDV), left ventricular end-systolic volume(LVESV), left ventricular internal diameter at end-diastole(LVIDd), and left ventricular internal diameter at end-systole(LVIDs). Left ventricular fractional shortening(LVFS) and left ventricular ejection fraction(LVEF) were also calculated. Four weeks after LAD ligation, blood samples were collected for serum biochemical measurements of serum creatinine(SCr) and blood urea nitrogen(BUN).Results Two weeks post-surgery, compared to the sham surgery group, the MI group showed increased LVESV and LVESD(P< 0.05), and decreased LVEF and LVFS(P< 0.001). No significant differences were observed in the MI+Veri group(P>0.05). Four weeks post-surgery, compared to the sham surgery group, both the MI and MI+Veri groups exhibited reduced LVEF and LVFS(P< 0.001). The MI+Veri group showed significantly lower LVESV and LVESD(P< 0.05) and higher LVEF and LVFS(P<0.001) compared to the MI group. Biochemical analysis of serum BUN levels indicated that the MI group had elevated BUN levels compared to the sham surgery group(P< 0.05), while the MI+Veri group showed lower BUN levels compared to the MI group(P< 0.05).Conclusions Vericiguat can partially improve acute heart failure and reverse ventricular remodeling in mice with acute myocardial infarction.
