贯叶金丝桃化学成分的分离与鉴定

【摘要】 目的 研究藤黄科(Guttiferae)金丝桃属(Hypericum)贯叶金丝桃(Hypericum perforatum L.)的化学成分。方法 采用正相硅胶色谱、反相ODS色谱、凝胶色谱及高效液相色谱等多种色谱方法对贯叶金丝桃体积分数95%乙醇提取物进行分离纯化。利用紫外、红外、核磁共振光谱,高分辨质谱及X-ray单晶衍射法对化合物的结构进行鉴定。结果 从贯叶金丝桃中分离鉴定8个化合物,分别为perforxanthone A(1)、8-hydroxy-1,2,3-trimethoxyxanthone(2)、绵马酚D(3)、(S)-5-hydroxy-3,4-dimethyl-5-pentylfuran-2(5H)-one(4)、ent-4(15)-eudesmene-1β,6α-diol(5)、methyl-(E)-8-oxooctadec-9-enoate(6)、模绕酮酸(7)、白桦脂酸(8)。结论 化合物1为新的氧杂蒽酮类化合物,化合物2-7为首次从该植物中分离得到。

【Abstract】 Objective To study the chemical composition of Hypericum perforatum L.Methods Various chromatographic methods such as normal-phase silica gel chromatography, reversed-phase silica gel chromatography, high performance liquid chromatography and gel chromatography were used to separate and purify the 95% EtOH extract of this plant.Structures of the isolated compounds were identified by NMR,UV,and IR spectral, high resolution mass spectrum(HRM),and X-ray single crystal diffraction.Results Eight compounds were isolated and identified from this plant, namely: perforxanthone A(1),8-hydroxy-1,2,3-trimethoxyxanthone(2),aspidinol D(3),(S)-5-hydroxy-3,4-dimethyl-5-pentylfuran-2(5H)-one(4),ent-4(15)-eudesmene-1β,6α-diol(5),methyl-(E)-8-oxooctadec-9-enoate(6),moronic acid(7),betulinic acid(8).Conclusion Compound 1 is a new compound.Compounds 2-7 are isolated from this plant for the first time.
