环总科线虫1种中国新纪录种的描述
Taxonomic of One New Record Species of Criconematoidea from China
【摘要】 环总科线虫是一类具有环型食道、根外寄生(少数为半内寄生)植物寄生性线虫,其寄主范围广,主要生活在农作物与木本植物根系周围。从中国浙江省杭州市女贞根围土壤中分离获得1种环总科线虫种群,经形态学和分子生物学鉴定,结果表明:分离到的环总科线虫为腓尼基半轮线虫(Hemicriconemoides phoenicis)。雌虫体长501.2 (439.2~519.6)μm,环形食道,有两个唇环,口针基球呈锚形,长为82.3 (79.8~85.2)μm,受精囊呈椭圆形且充满圆形的精子细胞,尾长29.8 (28.4~32.7)μm,幼虫和雌虫形态相似,雄虫未见。利用最大似然法(maximum likelihood, ML)对该线虫18S rDNA基因构建系统发育进化树,表明该线虫种群和H. phoenicis (MN660076)环总科半轮属线虫群体聚类在最高置信度(100%)的分支上。H. phoenicis是我国迄今首次报道的新纪录种,枣椰树是其首选寄主植物,女贞也是其寄主,拓宽了H. phoenicis的寄主范围。
【Abstract】 Criconematoidea were a class of parasitic nematodes with ring-type esophagus, extra-root parasitism(a few semiparasitic) plants, with a particularly wide range of hosts, mainly living around the roots of crops and woody plants. In this study, a new record species of Criconematoidea was isolated from privet rhizosphere soil in Hangzhou Zhejiang province,China. The morphological characteristics of the nematode were identified by optical microscopy. The results showed that the isolated nematode was identified as Hemicriconemoides phoenicis, a new record species in China. The body length of female Hemicriconemoides phoenicis is 501.2(439.2-519.6) μm, has a ring-shaped esophagus, two lip rings, and its knobs anchorshaped 82.3(79.8-85.2) μm long, spermatocyst is oval and filled with rounded sperm cells, a tail of 29.8(28.4-32.7) μm long. The juvenile is similar to the female. The male has not been seen. The maximum Likelihood method was used to construct a phylogenetic tree for the 18S rRNA gene of this nematode. The results showed that the isolated nematode were located on the branch of H. phoenicis(MN660076) cluster with the highest confidence(100%). H. phoenicis is a new record species reported for the first time in China. The date palm was the preferred host plant, but this study broadened the host range of H. phoenicis, suggesting that Ligustrum might also be its host plant.
