食管癌患者血清外泌体通过Wnt/β-catenin和PI3K-AKT信号通路促进癌细胞增殖及迁移
The serum exosomes of patients with esophageal cancer promote the proliferation and migration of cancer cells through Wnt/β-catenin and PI3K-AKT signaling pathways
【摘要】 目的:采用食管癌患者外周血来源的外泌体干预食管癌ECA109细胞,检测肿瘤细胞的增殖、凋亡、迁移及Wnt/β-catenin和PI3K-AKT信号通路蛋白的表达变化,探讨食管癌外泌体在肿瘤细胞发展进程中的作用。方法:超高速离心法提取食管癌患者外周血血清中的外泌体,与食管癌ECA109细胞共培养,检测细胞增殖、凋亡、迁移等生物学行为的变化;Western-blot检测AKT、β-catenin蛋白的表达。结果:透射电镜、粒径测定、标志蛋白结果证实提取的食管癌患者血清外泌体符合其鉴定标准;与空白细胞组相比,食管癌外泌体干预后食管癌ECA109细胞增殖、迁移能力显著提高,G2+S期细胞比例升高,细胞凋亡比例降低,统计学差异显著;食管癌外泌体组AKT蛋白磷酸化表达水平较空白细胞组明显升高,β-catenin蛋白表达显著降低。结论:食管癌来源的外泌体具有促进肿瘤细胞增殖与迁移、抑制细胞凋亡的作用,其机制可能与PI3K-AKT信号通路关键激酶AKT的激活有关,提示在肿瘤发展进程中肿瘤来源外泌体的作用不容忽视。
【Abstract】 Objective:To detect the proliferation, apoptosis and migration of tumor cells and the expression of Wnt/β-catenin and PI3K-AKT signaling pathways proteins of ECA109 cells by using exosomes derived from esophageal cancer, and to explore the role of exosomes in the development of esophageal cancer cells.Methods:Exosomes were extracted from peripheral blood serum of patients with esophageal cancer by ultra-high speed centrifugation method and co-cultured with ECA109 cells to detect the changes of cell proliferation, apoptosis, migration and other biological behaviors.The expressions of AKT and β-catenin were detected by Western-blot.Results:Transmission electron microscopy, particle size measurement and marker protein results confirmed that the extracted serum exosomes from patients with esophageal cancer met the identification criteria.Compared with the blank cell group, after the intervention of exosomes, the proliferation, migration and G2+S phase of esophageal cancer cells were significantly increased, and the apoptosis rate was significantly decreased.The phosphorylation level of AKT protein in the exosome group was significantly higher than that in the blank cell group, and the expression of β-catenin was significantly lower.Conclusion:Exosomes derived from esophageal cancer can promote the activation of AKT,promote the proliferation and migration of tumor cells, and inhibit cell apoptosis, suggesting that the role of tumor-derived exosomes in tumor development should not be ignored.
