高良姜素通过调节Hippo信号通路抑制甲状腺癌细胞的增殖、迁移和侵袭

【摘要】 目的:研究高良姜素(Galangin)对甲状腺癌细胞增殖、迁移和侵袭的影响以及Hippo信号通路发挥的作用。方法:培养人甲状腺癌细胞8505C,CCK-8实验检测不同浓度Galangin处理8505C细胞后,对8505C细胞存活率的影响,筛选实验用Galangin浓度;将8505C细胞分为对照组、Galangin组、Galangin+DMSO组、Galangin+XMU-MP-1组;EDU法检测各组8505C细胞增殖;Transwell检测各组8505C细胞侵袭;划痕实验检测各组8505C细胞迁移;流式细胞术检测各组8505C细胞凋亡率;RT-qPCR实验检测各组8505C细胞MST1、YAP、TAZ mRNA水平;Western blot实验检测各组8505C细胞MST1、YAP、TAZ、MMP2、Ki67、cleaved-Caspase3、Caspase3蛋白水平。结果:与对照组相比较,Galangin组8505C细胞EDU阳性细胞率、侵袭细胞数量、划痕愈合率、YAP、TAZ mRNA和蛋白水平、MMP2、Ki67蛋白水平显著性降低,细胞凋亡率、MST1 mRNA水平、MST1、cleaved-Caspase3/Caspase3蛋白表达水平显著性升高(P<0.05);与Galangin+DMSO组相比,Galangin+XMU-MP-1组8505C细胞EDU阳性细胞率、侵袭细胞数量、划痕愈合率、YAP、TAZ mRNA和蛋白水平、MMP2、Ki67蛋白水平显著性升高,凋亡率、MST1 mRNA水平、MST1、cleaved-Caspase3/Caspase3蛋白表达水平显著性降低(P<0.05)。结论:Galangin可能通过激活Hippo信号通路抑制甲状腺癌细胞8505C的增殖、迁移和侵袭。

【Abstract】 Objective:To study the impacts of Galangin on the proliferation, migration and invasion of thyroid cancer cells and the role of Hippo signaling pathway.Methods:Human thyroid cancer cell line 8505C was cultured.CCK-8 assay was used to detect the effect of different concentrations of Galangin on the survival rate of 8505C cells to screen the concentration of Galangin for experiment.8505C cells were divided into control group, Galangin group, Galangin+DMSO group, Galangin+XMU-MP-1 group.EDU method was applied to detect the proliferation of 8505C cells in each group.Transwell was applied to detect 8505C cell invasion in each group.Scratch test was used to detect 8505C cell migration in each group.Flow cytometry was applied to detect the apoptosis rate of 8505C cells in each group.RT-qPCR experiment was applied to detect the mRNA levels of MST1,YAP,and TAZ in 8505C cells in each group.Western blot experiment was applied to detect the protein levels of MST1,YAP,TAZ,MMP2,Ki67,cleaved-Caspase3,Caspase3 in 8505C cells in each group.Results:Compared with the control group, the rate of EDU positive cells, the number of invasive cells, scratch healing rate, the levels of YAP and TAZ mRNAs and proteins, the levels of MMP2,Ki67 proteins of 8505C cells in the Galangin group were obviously decreased, the apoptosis rate, the levels of MST1 mRNA and the levels of MST1,cleaved-Caspase3/Caspase3 protein were obviously increased(P<0.05).Compared with the Galangin+DMSO group, the rate of EDU positive cells, the number of invasive cells, scratch healing rate, the levels of YAP and TAZ mRNAs and proteins, the levels of MMP2,Ki67 proteins of 8505C cells in the Galangin+XMU-MP-1 group were obviously increased, the apoptosis rate, the levels of MST1 mRNA and the levels of MST1,cleaved-Caspase3/Caspase3 protein were obviously reduced(P<0.05).Conclusion:Galangin may inhibit the proliferation, migration and invasion of thyroid cancer cell line 8505C by activating Hippo signaling pathway.
