水产品中组胺快速检测的方法研究

【摘要】 目的:建立水产品中组胺快速检测的方法,并进行方法学验证。方法:本研究以偶氮试剂法-比色卡法作为水产品中组胺的快速检测手段。样品中的组胺经提取后,通过正戊醇萃取净化,再与偶氮试剂反应生成橙色化合物,其溶液颜色的深浅在一定范围内与样品中组胺的含量呈正相关,通过组胺标准色阶卡进行目视比色,对样品中组胺进行定性判定。结果:以鲈鱼、秋刀鱼、罗氏虾作为空白基质的检出限为200 mg·kg-1,灵敏度和特异性为100%、假阴性率和假阳性率为0%。结论:该方法快速准确,灵敏度高,适用于水产品中组胺的快速检测。

【Abstract】 Objective: To establish a rapid method for detecting histamine in aquatic products and validate it through methodological validation. Method: This study used the azo reagent colorimetric card method as a rapid detection method for histamine in aquatic products. After extraction, histamine in the sample is purified by n-pentanol extraction, and then reacts with azo reagents to generate orange compounds. The color depth of the solution is positively correlated with the content of histamine in the sample within a certain range. The histamine in the sample is qualitatively determined by visual colorimetry using the histamine standard color scale card. Result: The detection limit using sea bass, saury, and shrimp as blank matrices was 200 mg·kg-1, with a sensitivity and specificity of 100%and a false negative and false positive rate of 0%. Conclusion: This method is fast, accurate, highly sensitive, and suitable for the rapid detection of histamine in aquatic products.
