HPLC法测定天然植物饲料原料中菊苣酸和咖啡酸的含量

【摘要】 本实验旨在建立高效液相色谱法(HPLC)测定不同天然植物饲料原料中菊苣酸和咖啡酸含量的检测方法。样品经60%乙醇提取,使用Agilent 5 HC-C18色谱柱(250 mm×4.6 mm×5.0μm)分离,流动相甲醇/0.3%甲酸水,色谱柱温度为35℃,波长为327 nm,流速为0.8 mL/min,梯度洗脱,进样量为10μL。依照此法测定不同天然植物饲料原料中菊苣酸和咖啡酸的含量。结果表明:咖啡酸、菊苣酸在0.5~100.0μg/mL内线性关系良好(r>0.99),方法的检出限和定量限分别为0.6μg/mL和2μg/mL;供试样品在24 h内稳定性良好;平均加标回收率为81.1%~117.2%,相对标准偏差≤6.1%。该方法分离度好,精密度、稳定性均良好,加标回收率高,适用于天然植物饲料原料中菊苣酸和咖啡酸的测定。

【Abstract】 An high performance liquid chromatography(HPLC) method was developed for the simultaneous determination of chicorionic acid and caffeic acid in natural plant feedstuff. The sample was extracted with 60% ethanol,and separated by Agilent 5 HC-C18 column(250 mm×4.6 mm×5.0 μm). The mobile phase was methanol/0.3% formic acid,and the column temperature was 35 ℃. The detection wavelength was 327 nm, and the flow rate was 0.8 mL/min, gradient elution, and the injection volume was 10 μL. The contents of chicorionic acid and caffeic acid in different natural plant feedstuff were determined by this method. The results showed that there was a good linear relationship between the chicorionic acid and caffeic acid in a range of 0.5~100.0 ng/mL(r > 0.99). The limit of detection of the eight drugs was 0.6 μg/mL, and the limit of quantitation was 2 μg/mL. The test sample was stable within 24 hours, and the recoveries were 81.1%~117.2%, and the relative standard deviation was less than 6.1%. This method had good separation accuracy, precision, stability, and high recovery rate. It was suitable for the determination of chicory acid and caffeic acid in natural plant feedstuff.
